摘 要:目的探究雷公藤多苷片对免疫球蛋白A(IgA)肾病大鼠肾病理损伤及血清炎性因子表达的影响。方法用胎牛血清白蛋白刺激激活巨噬细胞的方法构建IgA肾病大鼠模型。将模型大鼠随机分为模型组和实验组,每组20只;另取20只正常大鼠作为对照组。对照组和模型组均予以等量0.9%NaCl;实验组按20 mg·kg^-1的剂量予以10 mg·mL^-1雷公藤多苷溶液。3组大鼠每天灌胃给药1次,连续给药4周。用苏木精-伊红染色检测大鼠肾组织病理学形态,用自动生化分析仪检测大鼠血清中肌酸酐(Cr)和尿素氮(BUN)水平,用酶联免疫吸附实验法测定大鼠血清中白细胞介素-6(IL-6)和IL-1β的含量。结果干预结束后,实验组、模型组和对照组的平均肾小球面积分别为(6.58±0.74)×10^3,(9.21±0.58)×10^3和(5.39±0.45)×10^3μm^2,平均肾小球体积分别为(6.45±0.74)×10^3μm^3,(8.69±1.05)×10^3μm^3和(4.89±0.46)×10^3μm^3,Cr分别为(38.55±3.75),(52.64±3.38)和(27.16±5.47)μmol·L^-1,BUN分别为(15.65±4.25),(23.54±5.26)和(7.87±2.31)μmol·L^-1,IL-6分别为(112.65±3.32),(143.48±4.37)和(19.44±5.27)pg·mg^-1,IL-1β分别为(36.55±4.43),(45.52±5.27)和(18.85±4.47)pg·mg^-1,实验组和对照组的上述指标和模型组比较,差异均有统计学意义(均P<0.05)。结论雷公藤多苷片能有效地改善IgA肾病大鼠的肾功能,降低血清炎性因子含量,减轻肾组织病理损伤。Objective To investigate the effects of tripterygium glycosides tablets on renal pathological damage and serum inflammatory factors expression in immunoglobulin A(IgA)nephropathy rats.Methods A rat model of IgA nephropathy was constructed by using fetal bovine serum albumin to stimulate activated macrophages.The model rats were randomly divided into model group and experimental group,with 20 rats in each group;another 20 normal rats were taken as the control group.Control group and the model group were given an equal amount of 0.9%NaCl.Experimental group was given 10 mg·mL^-1 tripterygium glycosides solution at a dose of 20 mg·kg^-1.Rats in the three groups were administered orally once a day for 4 weeks.Hematoxylin-eosin staining was used to detect the pathological morphology of rat kidney tissues,and the levels of creatinine(Cr)and brain urea nitrogen(BUN)in rat serum were measured using automatic biochemical analyzer.Interleukin-6(IL-6)and IL-1β levels in serum were measured using enzyme-linked immunosorbent assay.Results After intervention,the average glomerular area of the experimental group,model group and control group were(6.58±0.74)×10^3μm^3,(9.21±0.58)×10^3μm^3 and(5.39±0.45)×10^3μm^2,the average glomerular volume were(6.45±0.74)×10^3,(8.69±1.05)×10^3 and(4.89±0.46)×10^3μm^2,Cr were(38.55±3.75),(52.64±3.38)and(27.16±5.47)μmol·L-1,BUN were(15.65±4.25),(23.54±5.26)and(7.87±2.31)μmol·L^-1,IL-6 were(112.65±3.32),(143.48±4.37)and(19.44±5.27)pg·mg^-1,IL-1β were(36.55±4.43),(45.52±5.27)and(18.85±4.47)pg·mg^-1.The above indexes of experimental group and control group were compared with those of model group,and the differences were statistically significant(all P<0.05).Conclusion Tripterygium glycosides tablets can effectively improve the renal function of IgA nephropathy rats,reduce the content of serum inflammatory factors,and reduce the pathological damage of renal tissue.
