摘 要:目的研究异氟醚对宫颈癌裸鼠及小鼠宫颈癌U14细胞增殖、凋亡的影响。方法用HeLa细胞构建宫颈癌裸鼠模型,并随机分为实验组(1.4%异氟醚吸入2 h,同时在100%氧气条件下作用2 h)和对照组(在100%氧气下作用2 h),每组15只。2组裸鼠每天给药1次,连续干预10 d。另将U14细胞分为实验组(含1.4%异氟醚+5%CO2的气体环境中培养6 h)和对照组(含5%CO2的气体环境中培养6 h)。比较2组裸鼠的瘤重和存活期,以及2组U14细胞的存活率和克隆形成数。结果实验组和对照组的宫颈癌裸鼠瘤重分别为(1.69±0.15)和(2.73±0.16)g,存活期分别为(20.73±1.57)和(12.73±0.93)d,差异均有统计学意义(均P<0.05)。实验组和对照组的U14细胞存活率分别为(58.94±7.88)%和(100.00±6.79)%,克隆形成数分别为(52.67±4.19)和(89.33±5.73),差异均有统计学意义(均P<0.05)。结论异氟醚对宫颈癌裸鼠和细胞具有潜在的抑制作用。Objective To investigate the effect of isoflurane on the proliferation and apoptosis of cervical cancer in nude mice and cervical cancer U14 cells.Methods HeLa cells were used to construct a cervical cancer nude mouse model,and the mice were randomly divided into experimental group(1.4% isoflurane inhalation for 2 h,while acting under 100% oxygen for 2 h)and control group(acting under 100% oxygen for 2 h)with 15 cases per group.The nude mice of two groups were administered once a day for 10 days.In addition,U14 cells were divided into experimental group(1.4% isoflurane+5% CO2 in a gaseous environment for 6 h)and control group(5% CO2 in a gaseous environment for 6 h).The tumor weight and survival time of nude mice were compared between two groups.The survival rate and the number of clone formation of U14 cells were compared between two groups.Results The tumor weight of cervical cancer nude mice in experimental and control groups were(1.69±0.15)and(2.73±0.16)g,survival periods were(20.73±1.57)and(12.73±0.93)days,the differences were statistically significant(all P<0.05).The survival rates of U14 cells in experimental and control groups were(58.94±7.88)% and(100.00±6.79)%,the number of clone formations were(52.67±4.19)and(89.33±5.73),the differences were statistically significant(all P<0.05).Conclusion Isoflurane has a potential inhibitory effect on cervical cancer nude mice and cells.
