摘  要:目的研究槲皮苷在帕金森病(PD)细胞模型中对PC12细胞的保护作用及其机制。方法通过6-羟基多巴胺氢溴酸盐(6-OHDA)处理PC12细胞构建PD细胞模型。实验分为A,B,C和D组。A组加入0μmol·L^-16-OHDA和0μmol·L^-1槲皮苷;B组加入200μmol·L^-16-OHDA和0μmol·L^-1槲皮苷;C组加入200μmol·L^-16-OHDA和100μmol·L^-1槲皮苷;D组加入200μmol·L^-16-OHDA和200μmol·L^-1槲皮苷。用噻唑蓝法研究槲皮苷对6-OHDA处理后PC12细胞活性的影响,用TUNEL实验研究槲皮苷对细胞凋亡的影响,用蛋白质免疫印迹法研究槲皮苷对细胞磷脂酰肌醇3激酶(PI3K)/蛋白激酶B(Akt)/叉头转录因子O亚型3a(FoxO3a)信号通路的影响。结果槲皮苷可显著增加6-OHDA处理后PC12细胞活性,降低PC12细胞凋亡率,激活PI3K/Akt/FoxO3a信号通路并抑制细胞凋亡通路。结论槲皮苷通过激活PI3K/Akt/FoxO3a信号通路,提高6-OHDA处理后神经细胞活性,降低凋亡水平,为神经细胞提供保护。Objective To study the protective effect and mechanism of quercetin on PC12 cells in Parkinson’s disease(PD)cell model.Methods The PD model of PC12 cells treated with 6-hydroxydopamine hydrobromide(6-OHDA)was constructed.The experiment is divided into A,B,C and D groups.A group was given 0μmol·L^-16-OHDA and 0μmol·L^-1 quercetin.B group was given 200μmol·L^-16-OHDA and 0μmol·L^-1 quercetin.C group was given 200μmol·L^-16-OHDA and 100μmol·L^-1 quercetin.D group was given 200μmol·L-16-OHDA and 200μmol·L^-1 quercetin.Methyl thiazolyl tetrazolium experiment was used to study the effect of quercetin on the activity of PC12 cells after 6-OHDA treatment.TUNEL experiment was conducted to study the effect of quercetin on cell apoptosis.The effect of quercetin on cellular phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)/forkhead transcription factor O subtype 3a(FoxO3a)signaling pathway was investigated by Western Blotting.Results Quercetin can significantly increase the activity of PC12 cells after 6-OHDA treatment,reduce the apoptosis rate of PC12 cells,activate PI3K/Akt/FoxO3a signaling pathway and inhibit the apoptosis pathway.Conclusion Quercetin provide a protection for nerve cells by increasing the activity of nerve cells after 6-OHDA treatment and decreasing the apoptotic level through activating PI3K/Akt/FoxO3a signaling pathway.
