摘 要:目的探讨大鼠间充质干细胞(mesenchymal stem cell,MSCs)对脓毒症性急性肺损伤(sepsis induced acute lung injury,SI-ALI)大鼠肺内皮细胞(endothelial cells,ECs)再生能力的作用。方法饲养33只成年雄性SD大鼠,3只用于获取MSCs;另外30只大鼠被随机划分为对照组、SI-ALI组和SI-ALI+MSCs组,每组10只。利用细菌脂多糖(lipopolysaccharide,LPS)腹腔注射的方法构建SI-ALI模型,MSCs在构建模型后的2小时内通过尾静脉注射入大鼠体内,并在模型构建72小时后获取各组大鼠的肺组织。利用逆转录-聚合酶链反应(reverse transcriotion-polymerase chain reaction,RT-PCR)检测各组大鼠肺组织中炎症因子IL-1、IL-6和TNF-α的mRNA表达水平,利用流式细胞术(flow cytometry,FCM)检测肺组织中ECs的再生能力,并计算各组大鼠的存活率。结果与对照组相比,SI-ALI组大鼠肺组织中炎症因子IL-1、IL-6和TNF-α的表达水平升高,ECs的再生能力降低,大鼠的总体存活率下降。与SI-ALI组相比,SI-ALI+MSCs组大鼠肺组织中炎症因子IL-1、IL-6和TNF-α的表达水平降低,ECs的再生能力增强,大鼠的总体存活率升高。结论间充质干细胞抑制了SI-ALI大鼠肺组织中炎症反应、增强了ECs的再生能力,并提高了存活比率。Objective To investigate the effect of mesenchymal stem cells(MSCs)on the regeneration ability of endothelial cells(ECs)in rat with sepsis induced acute lung injury(SI-ALI).Methods A total of 33 male SD rats were selected in this study,3 of them were used to obtain MSCs,and the other 30 rats were randomly divided into three groups:the control group,the SI-ALI group and the SI-ALI+MSCs group,with 10 rats in each group.The SI-ALI rat model was constructed through lipopolysaccharide(LPS)intraperitoneal injection.MSCs were administrated into rats through tail vein injection,and the lung sample was gathered at 72 hours after LPS injection.Reverse transcription polymerase chain reaction(RT-PCR)was performed to measure the expression of cytokines IL-1,IL-6 and TNF-αin lung tissues.Flow cytometry(FCM)was performed to measure the regeneration ability of ECs in lung.The survival rate of rats in each group was counted as well.Results Compared with the control group,the expression of cytokines IL-1,IL-6 and TNF-αin lung tissues of the SI-ALI group increased,the regeneration ability of ECs in the lung decreased,and the total survival rate of rats decreased.Compared with the SI-ALI group,the expression of cytokines IL-1,IL-6 and TNF-αin lung tissues of the SI-ALI+MSCs group decreased,the regeneration ability of ECs in lung increased,and the total survival rate of rats increased as well.Conclusion MSCs can inhibit the inflammation reaction in the lung of SI-ALI rats,enhance the regeneration ability of ECs and increase the total survival rate.
