摘  要:目的:研究墨旱莲对高脂喂养小鼠肝脏脂代谢的影响。方法:选取70只雌性C57BL/6J小鼠,按随机数字表法随机分为对照组、高脂模型组和低、中、高剂量墨旱莲给药组。对照组小鼠喂饲基础饲料,高脂模型组和低、中、高剂量墨旱莲给药组小鼠喂饲高脂饲料。对照组、高脂模型组小鼠灌胃给予1%羧甲基纤维素钠(CMC-Na)溶液,低、中、高剂量墨旱莲给药组小鼠灌胃给予1%CMC-Na溶液配制的墨旱莲灌胃液40、120、360 mg/kg,每天给药1次,共6周,第6周测定小鼠体质量、Lee’s指数、体质量指数(BMI),血清中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、三酰甘油(TG)、总胆固醇(TC)、葡萄糖(Glu)的水平,并解剖分离小鼠肝脏,测定肝脏重量、肝脏重量与体质量比值、腹部及肾周脂肪重量、脂肪重量与体质量比值,测定游离脂肪酸(FFA)含量并制作苏木精-伊红(HE)染色切片。结果:给药6周后,与对照组比较,高脂模型组小鼠体质量、Lee’s指数、BMI、TC、TG均显著升高(P<0.05),肝脏重量、肝脏重量与体质量比值、腹部及肾周脂肪重量、脂肪重量与体质量比值均有显著改变(P<0.05)。与高脂模型组比较,中、高剂量墨旱莲给药组小鼠体质量、Lee’s指数、BMI、腹部及肾周脂肪重量、肝脏重量与体质量比值均显著降低(P<0.05),高剂量墨旱莲给药组小鼠TC、TG显著降低(P<0.05)。在肝脏HE切片中,高脂模型组小鼠可见肝脏有大量脂肪空泡,而对照组和高剂量墨旱莲给药组未见肝细胞脂肪变性。结论:墨旱莲能改善高脂喂养小鼠的肝脏脂代谢。Objective:To investigate the effects of Ecliptae Herba on liver lipid metabolism in mice fed with high-fat diet.Methods:Seventy female C57BL/6J mice were randomly divided into five groups:the control group,the high-fat model group,the low,medium and high-dose Ecliptae Herba groups.The mice in the control group were fed with basal diet,and the mice in the other groups were fed with high-fat diet.Meanwhile,the mice in the control group and the high-fat model group were gavaged with 1%carboxy methyl cellulose sodium(CMC-Na)solution,and the mice in the low,medium and high-dose Ecliptae Herba groups were given 40,120,360 mg/kg Ediptae Herba solution prepared with 1%CMC-Na by gavage,once a day for a succession of 6 weeks.After 6 weeks,the body weight,Lee’s index,body mass index(BMI),the levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),triglyceride(TG),total cholesterol(TC)and glucose(Glu)were determined.Liver tissues were separated,liver weight,liver weight/body weight ratio,abdominal and perirenal fat weight,fat weight/body weight ratio were detected and calculated,and the content of free fatty acid(FFA)was determined.Hematoxylin-eosin(HE)staining sections of the liver tissues were also prepared.Results:After 6 weeks of medication,the levels of body weight,Lee’s index,BMI,TC and TG of the mice in the high-fat model group increased significantly,as compared with those of the control group(P<0.05),while the liver weight,liver weight/body weight ratio,abdominal and perirenal fat weight,fat weight/body weight ratio of the mice in the high-fat model group displayed significant changes(P<0.05).Compared with the high-fat model group,the body weight,Lee’s index,BMI,abdominal and perirenal fat weight,liver weight/body weight ratio of the mice in the medium and high-dose Ecliptae Herba groups decreased significantly(P<0.05),and TC and TG in the high-dose Ecliptae Herba group also significantly decreased(P<0.05).Many fat empty bubbles could be seen in the liver HE sections of the high-fat model group
