摘  要:目的探究姜黄素促进Syndecan-1(SDC1)表达,调控细胞紧密连接,对严重腹腔感染时肠屏障功能的保护作用。方法选取由广州医科大学实验动物中心提供120只基因敲除大鼠作为研究对象,根据随机数字表法分为空白对照组、对照组和治疗组各40只。空白对照组只剖腹,对照组行盲肠结扎术,治疗组在盲肠结扎术后静脉滴注姜黄素,检测各组SDC1蛋白表达、核因子kB(NF-kB)蛋白表达、mRNA水平和以及趋化因子10、白细胞介素8、肿瘤坏死因子α等炎性因子水平。结果治疗组SDC1蛋白表达、NF-kB蛋白表达和mRNA水平低于对照组和空白对照组,对照组SDC1蛋白表达、NF-kB蛋白表达和mRNA水平低于空白对照组,差异有统计学意义(P<0.05);治疗组IP-10、IL-8、TNF-α水平低于对照组和空白对照组,对照组IP-10、IL-8、TNF-α水平低于空白对照组,差异均有统计学意义(P<0.05)。结论姜黄素可以促进SDC1表达,调控细胞的紧密连接,形成对严重腹腔感染时屏障功能的保护。Objective To explore the protective effect of curcumin on intestinal barrier function in severe abdominalinfection by promoting the level of syndecan-1 andregulation of tight cell connections. Methods 120 knockout rats were selected from theexperimental animal center of Guangzhou Medical University, and were divided into blank control group, control group andtreatment group by means of random number table. There were 40 rats in each group. The blank control group was only given the laparotomy, the control group was givencecal ligation, and the treatment group was received intravenous infusion of curcumin after cecal ligation. Protein expression and mRNA levels of SDC1, nuclear factor kB and levels of inflammatory factors such as chemokine 10, interleukin 8, and tumor necrosis factor alpha were detected in each group. Results Protein expression and mRNA level of SDC1, NF-KB in the treatment group were lower than those in the control group and the blank control group, andthe differences were statistically significant(P<0.05). The levels of IP-10, IL-8 and TNF-in the treatment group were lowerthan those in the control group and the blank control group, and the levels of IP-10, IL-8 and TNF-in the control group werelower than those in the blank control group, with statistically significant differences(P<0.05). Conclusion Curcumin canpromote the expression of SDC1 and regulate cell tight junctionto protect the barrier function in severe abdominal infection.
