摘 要:目的:优选黄连降脂合剂提取工艺条件。方法:建立合剂中葛根素含量的高效液相测定方法,基于药材浸润时间和溶剂用量、料液比、提取时间的单因素考察结果,设计L9(3)4正交试验,以葛根素含量和干膏收率的综合评分为指标,优选提取水量、时间、次数3个影响提取效果的工艺参数。结果:葛根素质量浓度在5.16~165.20μg·ml^-1范围内与吸光度呈良好的线性关系(r=1.0000);重复性试验、精密度试验的RSD均小于2%,平均加样回收率为100.63%(RSD为1.20%,n=9)。单因素考察结果显示,各因素对水提取工艺的影响顺序:提取次数﹥提取水量﹥提取时间;优选工艺条件为:加10倍量水,提取2次,每次2.0 h。葛根素平均转移率为73.72%。结论:优选的提取工艺稳定可行,适用于黄连降脂合剂的规范化生产。Objective: To optimize the extraction process of Huanglian Jiangzhi mixture. Methods: A method was established for the determination of puerarin in the mixture by HPLC. L9(3)4 orthogonal experiment was designed based on the single factor investigation results of soaking time,solvent dosage,material-liquid ratio and extraction time. Taking the comprehensive score as the index,three process parameters affecting the extraction effect were optimized. Results: The linear range of dendrobine was 5.16^-165.20 μg·ml^-1( r = 1.000 0). The RSDs of repeatability and precision tests were all lower than 2%. The average recovery was 100.63%( RSD = 1.20%,n = 9). The single factor inspection determined the suitable scopes of technological investigation for orthogonal experiment. The order of influencing factors on the water extraction process was extraction times,extraction water quantity and extraction duration. The optimal extraction condition was ten-fold amount of water,and extracting twice with 2 hours for each time. Conclusion: The optimal process is stable,and can be used for the preparation of Huanglian Jiangzhi mixture.
