摘 要:目的探讨羟基红花黄色素A对心肌缺血再灌注损伤大鼠血清炎性因子及心肌组织核因子-κB(NF-κB)表达的影响。方法将50只雄性SD大鼠随机分为假手术组、缺血再灌注组(模型组)、尼莫地平组(阳性药对照组,20 mg/kg)、羟基红花黄色素A高剂量组(20 mg/kg)、羟基红花黄色素A低剂量组(10 mg/kg),每组10只。所有大鼠均灌胃给药,每日1次。给药14 d后,以结扎SD大鼠冠状动脉左前降支建立心肌缺血再灌注损伤模型。各组大鼠缺血30 min再灌注120 min后,检测各组大鼠心脏功能,并采集颈总动脉血液,测定血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、C反应蛋白(CRP)等炎性因子,同时取大鼠心肌组织,光镜下观察心肌组织结构,并采用Western Blotting法检测心肌NF-κB、NF-κB抑制蛋白激酶α(IκBα)、NF-κB抑制蛋白激酶β(Iκκβ)等表达水平。结果与假手术组比较,模型组大鼠左室舒张末期压力(LVEDP)升高,左室收缩压(LVSP)降低,血清TNF-α、IL-6、CRP炎性因子水平升高,心肌组织NF-κB蛋白表达升高,IκBα、Iκκβ蛋白表达降低,差异均有统计学意义(P<0.05)。与模型组比较,羟基红花黄色素A高剂量组、低剂量组及尼莫地平组大鼠LVEDP降低,LVSP升高,血清TNF-α、IL-6、CRP炎性因子水平均降低,心肌组织NF-κB蛋白表达均降低,IκBα、Iκκβ蛋白表达均升高,差异均有统计学意义(P<0.05)。结论羟基红花黄色素A对心肌缺血再灌注损伤大鼠具有保护作用,其作用机制可能与调控炎性因子及NF-κB信号通路有关。Objective To investigate the effects of hydroxysafflor yellow A(HSYA)on inflammatory factors and expression of nuclear factor-κB(NF-κB)in rats with myocardial ischemia-reperfusion injury.Methods Fifty SD male rats were randomly divided into sham-operated group,ischemia reperfusion group(model group),nimodipine group(positive control group,nimodipine 20 mg/kg),HSYA high dose group(20 mg/kg),and hydroxysafflor yellow A(HSYA)low dose group(10 mg/kg),10 rats in each group.Each rat was gavaged once a day.After 14 days,the animal model of myocardial ischemia-reperfusion injury was established by ligating the left anterior descending coronary artery of SD rats.After 30 minutes of ischemia and 120 minutes of reperfusion,the cardiac function of rats in each group was measured,and tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),C-reactive protein(CRP)were detected.Meanwhile,myocardial tissue was observed by light microscopy,and the levels of NF-κB,NF-κB inhibitory protein kinaseα(IκBα),NF-κB inhibitory protein kinaseβ(Iκκβ)were detected by Western Blotting.Results Compared with the sham-operated group,the left ventricular end-diastolic pressure(LVEDP)and left ventricular systolic pressure(LVSP)were significantly increased,the levels of TNF-α,IL-6,and CRP were significantly increased,the expression of NF-κB in myocardial tissue was significantly increased,and the expression of IκBα,Iκκβprotein was significantly decreased in the model group(P<0.05).Compared with model group,the levels of LVEDP,LVSP,TNF-α,IL-6,and CRP were significantly decreased,the expression of NF-κB protein in myocardial tissue was significantly decreased,and the expressions of IκBα,Iκκβprotein were significantly increased in HSYA high dose group,HSYA low dose group,and positive control group(P<0.05).Conclusion Hydroxysafflor yellow A had protective effect in rats on myocardial ischemia-reperfusion injury,and its mechanism may be related to the regulation of inflammatory factors and NF-κB signaling pathway.
