慢粒灵制剂中青黛定性鉴别和红花3种黄酮类成分含量测定

摘　　要：目的建立鉴别慢粒灵颗粒(胶囊)中青黛的薄层色谱(TLC)法及同时测定红花中羟基红花黄色素A、6-羟基山柰酚-3-O-β-葡萄糖苷和红花黄色素B含量的超高效液相色谱(UPLC)法。方法采用TLC法鉴别青黛。色谱柱为ACQUITY UPLC BEH C18柱(100 mm×2.1 mm,1.7μm),流动相为乙腈-0.01%三氟乙酸水溶液,梯度洗脱,流速为0.3 m L/min,柱温为30℃,检测波长为380 nm,进样量为10μL。结果羟基红花黄色素A、6-羟基山柰酚-3-O-β-葡萄糖苷和红花黄色素B的进样量分别在25.04~5008.0 ng,21.36~4272.0 ng,19.92~3984.0 ng范围内与峰面积线性关系良好(r≥0.9999),平均回收率分别为96.43%,95.88%,95.25%,RSD分别为1.48%,0.66%,1.06%(n=6)。结论该方法分析速度快,灵敏度高,结果准确,可用于慢粒灵颗粒(胶囊)中青黛的鉴别和红花药材中3种黄酮类成分的含量测定。Objective To establish a thin layer chromatography(TLC)method for the identification of Indigo naturalis and ultra performance liquid chromatography(UPLC)method for the determination of hydroxy safflor yellow A(HSYA),6-hydroxy kaempferol-3-O-glucoside(6-HK3-O-G)and safflower yellow B(SYB)in Manliling Granules(Capsules).Methods The TLC method was used to identification of Indigo naturalis,the ACQUITY UPLC BEH C18 chromatographic column(100 mm×2.1 mm,1.7μm)was used with acetonitrile(A)-0.01%TFA(B)as the mobile phase of gradient elution at flow rate of 0.3 m L/min,detection wavelength was maintain at 380 nm,column temperature was maintained at 30℃and the injection volume was 10μL.Results Hydroxysafflor yellow A,6-hydroxykaempferol-3-O-glucoside and safflower yellow B showed good linear relationship with the peak area when the injection volume was in the ranges of 25.04-5008.0 ng,21.36 ng-4272.0 ng,and 19.92 ng-3984.0 ng(r≥0.9999),respectively.The average recoveries were 96.43%,95.88%and 95.25%within RSDs of 1.48%,0.66%and 1.06%(n=6).Conclusion The method is rapid,sensitive and reproducible,which can be used for qualitative identification of Indigo naturalis and determination of three flavones of Flos carthami in Manliling Granules(Capsules).
