摘 要:目的观察心痛泰颗粒对冠心病大鼠血清中血脂、纤溶系统的影响及心肌组织形态学改变,并探讨心痛泰对冠心病大鼠的治疗作用及机制。方法将95只4周龄SD大鼠随机抽取17只作为空白组。其余78只SD大鼠进行造模(灌胃高脂饲料12周),造模后随机抽取75只分为模型组、心痛泰高剂量组、心痛泰中剂量组、心痛泰低剂量组、卡托普利组,每组各15只。第13周开始,各组分别予相应药物灌胃治疗,正常组和模型组以等体积的蒸馏水灌胃,每日1次,连续灌胃4周。于实验后,采用腹主动脉取血,化学发光法检测甘油三酯(TAG)、胆固醇(TC)水平。使用ELISA法检测血浆纤溶酶原激活剂(t-PA)和纤溶酶原激活物抑制物(PAI-1)水平;取血结束后迅速活体取出心脏观察病理组织结构。结果1)心痛泰低剂量组与模型组比较TC降低(P<0.05),心痛泰高剂量组、心痛泰中剂量组与模型组比较TC均明显降低(P<0.01)。2)心痛泰低剂量组与模型组比较TAG降低(P<0.05),心痛泰高剂量组、心痛泰中剂量组与模型组比较TAG均明显降低(P<0.01)。3)模型组大鼠PAI-1高脂饲料喂养后明显高于空白组(P<0.01),模型组大鼠t-PA明显低于空白组(P<0.01)。4)心痛泰低剂量组与模型组比较,PAI-1降低(P<0.05),心痛泰高剂量组、心痛泰中剂量组、卡托普利组与模型组比较,PAI-1明显降低(P<0.01)。5)心痛泰低剂量组与模型组比较,t-PA升高(P<0.05),心痛泰高剂量组、心痛泰中剂量组、卡托普利组与模型组比较,t-PA明显升高(P<0.01)。6)心痛泰低剂量组:心肌纤维分布大致整齐,心肌细胞间质稍增宽,且未见显著炎性细胞浸润,水肿属于轻度。心痛泰中剂量组:心肌纤维排列整齐度一般,心肌细胞间质轻度增宽,且未见明显炎性细胞浸润,水肿属于轻度。心痛泰高剂量组:心肌纤维排列较前几组整齐,心肌细胞间质稍增宽,且未见明显炎性细胞浸润,水肿属于轻度。�Objective:To observe the effect of Xintongtai on serum lipids,fibrinolytic system and myocardial histomorphology in rats with coronary heart disease,and to explore its therapeutic effect and mechanism.Methods:17 out of 954-week-old SD rats were randomly selected as the blank control group,and the other 78 ones were modeled.75 rats were randomly divided into the model group,high-dose group,middle-dose group,low-dose group and Captopril group according to the weight stratification,15 rats in each.At the beginning of the 14 th week,each group was treated with corresponding drugs.The normal group and the model group were treated with equal volume distilled water once a day for 4 weeks.After the experiment,blood was collected from abdominal aorta;the levels of cholesterol and triglyceride were detected with chemiluminescence method;the levels of t-PA and PAI-1 were detected by ELISA,and the heart tissue structure was observed in vivo after blood collection.Results:1)compared with the model group,the cholesterol of xintongtai low dose group decreased significantly(P<0.05),the cholesterol of xintongtai high dose group,xintongtai middle dose group and the model group decreased significantly(P<0.01).2)compared with the model group,the triglyceride of all xintongtai groups decreased significantly(P<0.05 or P<0.01).3)Compared with the model group,the triglycerides in the high dose group and the middle dose group decreased significantly(P<0.01);the PAI-1 high-fat diet in the model group was significantly higher than that in the blank group(P<0.01),and the t-PA in the model group was significantly lower than that in the blank group(P<0.01).4)The PAI-1 of xintongtai low-dose group was lower than that of model group,the difference was statistically significant(P<0.05).PAI-1 of xintongtai high-dose group,xintongtai middle dose group and captopril group was significantly lower than that of model group,the difference was statistically significant(P<0.01).5)t-PA of xintongtai low-dose group was higher than that of model group,the d
