摘  要:目的研究虎杖、半枝莲、蝉蜕及其药对和小复方提取物对支原体肺炎(MPP)小鼠炎症因子及脂代谢信号分子的影响。方法雄性BALB/c小鼠90只,体质量(18±2)g,3周龄,随机分为9组:正常组、模型组、虎杖组、半枝莲组、蝉蜕组、虎杖+半枝莲组、虎杖+蝉蜕组、半枝莲+蝉蜕组和小复方组。正常组以生理盐水滴鼻,模型组和各治疗组以支原体(MP)菌液滴鼻,连续3 d。治疗从感染第1天开始,连续3 d。第4天处死小鼠,ELISA法检测小鼠肺泡灌洗液中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、cPLA2、消退素D1(RvD1)的水平;PCR法检测小鼠肺组织内MP-DNA的表达;WB检测小鼠肺组织中核转录因子-κB(NF-κB)、LOX-1、磷脂酶A2(PLA2)、保护素D1(PD1)的表达;HE染色观察小鼠肺组织的生理病理形态。结果与正常组相比,模型组BALF中TNF-α、IL-6、cPLA2的水平均明显升高(P<0.01),RvD1的水平明显降低(P<0.01),各治疗组相比模型组TNF-α、IL-6、cPLA2的水平均明显降低(P<0.01),RvD1的水平明显升高(P<0.01);与正常组相比,模型组小鼠肺组织中NF-κB、LOX-1、PLA2的表达均明显升高(P<0.01),PD1的表达明显降低(P<0.01),各治疗组相比模型组NF-κB、LOX-1、PLA2的表达均明显降低(P<0.01),PD1的表达明显升高(P<0.01);正常组小鼠肺组织结构基本完好,模型组小鼠的支气管周围和肺间质可见大量炎症细胞浸润,肺泡壁毛细血管扩张充盈,肺泡壁增厚破坏,支气管管腔变窄,各治疗组对肺组织炎性渗出和血管扩张情况均有不同程度的改善。结论虎杖、半枝莲、蝉蜕提取物可降低MPP小鼠TNF-α、IL-6的水平,抑制NF-κB活化,降低LOX-1、PLA2的水平,升高RvD1、PD1的水平,推测其治疗MPP可通过调节"炎症-脂代谢"相关靶点而起效。Objective:To investigate the effect of traditional Chinese medicine extracts on inflammatory factors and lipid metabolism signaling molecules in MPP mice.Methods:90 male BALB/c mice,body weight(18±2)g,3-week old,were randomly divided into 9 groups:the control group,the model group,Polygonum cuspidate group,Barbed skullcap group,Periostracum cicadae group,Polygonum cuspidate+Barbed skullcap group,Polygonum cuspidate+Periostracum cicadae group,Barbed skullcap+Periostracum cicadae group and formula group.The control group were treated with normal saline by nasal drip while the model group and the treatment groups were infected with MP by nasal drip for three consecutive days.Treatment began on the first day of the infection for three consecutive days.Mice were killed on the fourth day.ELISA was used to detect the levels of IL-6,TNF-α,c PLA2 and RvD1,PCR to measure the level of MP-DNA,WB to detect the expressions of NF-κB,LOX-1,PLA2 and PD1,HE to observe the physiological and pathological structure of lung tissue.Results:Compared with the normal group,the levels of TNF-α,IL-6 and cPLA2 in BALF of the model group were increased significantly(P<0.01),and the level of RvD1 was significantly decreased.Compared with the model group,levels of TNF-α,IL-6 and cPLA2 in each treatment group were significantly reduced(P<0.01),and the level of RvD1 was significantly increased(P<0.01).The expressions of NF-κB,LOX-1 and PLA2 in the model group were increased significantly compared with the normal group(P<0.01),but the expression of PD1 was decreased significantly(P<0.01).And the expressions of NF-κB,LOX-1 and PLA2 in treatment groups were significantly reduced(P<0.01),while the expression of PD1 was significantly increased(P<0.01).In the control group,the lung tissue was basically intact.A large number of inflammatory cells infiltrated around bronchi and pulmonary interstitial of the model group;alveolar wall was thickened even destructed and its capillaries were expanded,bronchial lumen narrowed.The inflammatory exudation
