摘 要:目的:研究Toll样受体4(toll-like receptors 4,TLR4)/核转录因子-κB(nuclear factor-kappa B,NF-κB)在口腔正常舌黏膜、颊囊黏膜癌前病变、口腔鳞癌表达情况及其意义。方法:以前期工作中收集的新鲜冻存的金黄地鼠口腔正常舌黏膜、颊囊黏膜癌前病变、口腔鳞癌作为实验对象,从各个层次,应用RT-PCR、实时荧光定量PCR、Western blot、ELISA等技术验证TLR4及NF-κB的mRNA及蛋白表达情况。结果:与口腔正常舌黏膜相比,TLR4及NF-κB的mRNA及蛋白在颊囊黏膜癌前病变、口腔鳞癌中逐渐升高,口腔鳞癌中升高尤为明显,相对于正常舌黏膜、颊囊黏膜癌前病变,差异有统计学意义(P<0.01)。结论:TLR/NF-κB信号通路在口腔鳞癌中被激活,可能与口腔鳞癌的发生发展有关,进一步阐明其在口腔鳞癌的病因和发病机制中的作用有重要意义,可能会为以后口腔鳞癌的防治提供依据。Objective:To investigate the expression of Toll-like receptor 4(TLR4)and nuclear factor-kappa B(NF-κB),as well as its significance,in the normal tongue mucosa,precancerous lesions of the cheek pouch mucosa,and oral squamous cell carcinoma in golden hamsters.Methods:We had prepared the fresh frozen tissues of the normal tongue mucosa,precancerous lesions of the cheek pouch mucosa,and oral squamous cell carcinoma in previous research.We determined the mRNA and protein expression levels of TLR4 and NF-κB in these tissues by RT-PCR,real-time PCR,Western blot,and ELISA.Results:Compared with those in the normal mucosa of the tongue,the mRNA and protein expression levels of TLR4 and NF-κB were increased in the precancerous lesions of the cheek pouch mucosa,and were further increased in oral squamous cell carcinoma;and the differences reached statistical significance between oral squamous cell carcinoma and the normal mucosa or precancerous lesions(P<0.01).Conclusion:The TLR4/NF-κB signaling pathway is activated in oral squamous cell carcinoma,possibly related to the development and progression of the cancer.It is of great significance to clarify the role of this pathway in the etiology and pathogenesis of oral squamous cell carcinoma,which may provide a basis for the prevention and treatment of oral squamous cell carcinoma.
