摘　　要：目的:建立多通道HPLC法同时测定龙胆泻肝胶囊中栀子苷、龙胆苦苷、黄芩苷、甘草酸铵的含量。方法:色谱柱:Kromasil 100-5 C18(125 mm×4.0 mm,5μm);流动相:乙腈(A)-0.1%磷酸(B),梯度洗脱;流速:1.0 mL/min;柱温:30℃;检测波长:238 nm(栀子苷)、270 nm(龙胆苦苷)、280 nm(黄芩苷)、250 nm(甘草酸铵);进样体积:10μL。结果:栀子苷、龙胆苦苷、黄芩苷、甘草酸铵分别在8.526~170.52、3.774~75.48、6.867~137.34、1.66~33.16μg/mL范围内与峰面积呈良好线性关系(r>0.999);精密度、稳定性、重复性良好,RSD均小于2%;平均回收率分别为101.08%(RSD=1.73%)、101.10%(RSD=1.81%)、98.48%(RSD=1.21%)、101.47%(RSD=1.34%)。结论:该方法简单、准确、快速,可用于龙胆泻肝胶囊的质量控制及后续质量标准的提高。Objective:To establish a method based on multichannel high-performance liquid chromatography(HPLC)for simultaneous determination of the content of gentiopicroside,geniposide,baicalin,and ammonium glycyrrhizinate in Longdanxiegan capsules.Methods:HPLC was performed on a Kromasil 100-5 C18 column(125 mm×4.0 mm,5μm)at 30℃using the mobile phase of acetonitrile(A)-0.1%phosphoric acid(B)for gradient elution at a flow rate of 1.0 mL/min.The detection wavelength was 238 nm for geniposide,270 nm for gentiopicroside,280 nm for baicalein,and 250 nm for ammonium glycyrrhizinate.The injection volume was 10μL.Results:There was a good linear relationship between concentration and peak area for geniposide within 8.526~170.52μg/mL,for gentiopicroside within 3.774~75.48μg/mL,for baicalin within 6.867~137.34μg/mL,and for ammonium glycyrrhizinate within 1.66~33.16μg/mL(r>0.999).The average recoveries were 101.08%(relative standard deviation[RSD]=1.73%),101.10%(RSD=1.81%),98.48%(RSD=1.21%),and 101.47%(RSD=1.34%),respectively.There was good precision,stability,and repeatability,with RSD less than 2%.Conclusion:The established method is simple,rapid,and accurate,and can be used for the quality control of Longdanxiegan capsules and subsequent improvement of quality standards.
