摘 要:目的:建立高效液相色谱(high-performance liquid chromatography, HPLC)同时测定月桂油中丁香酚、芳樟醇和桉油精含量的方法。方法:试验采用Dikma Diamonsil C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈:甲醇:水(35∶29∶36);检测波长200 nm;柱温30℃;流速1.0 mL/min。结果:丁香酚、芳樟醇和桉油精分别在0.021~0.208 mg/mL(r=0.999 9)、0.048~0.481 mg/mL(r=0.999 8)、0.679~6.788 mg/mL(r=0.999 5)范围内,与其峰面积呈较好的线性关系。加样回收率分别为100.34%、100.08%、99.45%,RSD分别为1.18%、1.32%、1.47%。测得三批月桂油中3种成分的含量分别为12.73~14.17mg/g、26.50~29.76 mg/g、415.99~459.63 mg/g。结论:该方法简便快速、准确灵敏,具有良好的重复性和回收率,可作为月桂油中丁香酚、芳樟醇和桉油精的定量分析方法,为月桂油的质量控制和评价提供科学依据。Objective: To simultaneously determine the content of eugenol, linalool, and eucalyptol in laurel oil by high-performance liquid chromatography(HPLC). Methods: HPLC was performed on a Dikma Diamonsil C18 column(250 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile-methanol-water(35∶29∶36) at a detection wavelength of 200 nm, a column temperature of 30 °C, and a flow rate of 1.0 mL/min. Results: The concentrations of eugenol, linalool, and eucalyptol in laurel oil showed good linear relationships with the corresponding peak areas in the ranges of 0.021 ~ 0.208 mg/mL(r = 0.9999), 0.048 ~ 0.481 mg/mL(r = 0.999 8), and 0.679 ~ 6.788 mg/mL(r =0.9995), respectively. The average recoveries of eugenol, linalool, and eucalyptol were 100.34 %(RSD [relative standard deviation] = 1.18 %), 100.08 %(RSD = 1.32 %), and 99.45 %(RSD = 1.47 %), respectively. The content of eugenol, linalool, and eucalyptol in three batches of laurel oil was 12.73 ~ 14.17 mg/g, 26.50 ~ 29.76 mg/g, and415.99 ~ 459.63 mg/g, respectively. Conclusion: The HPLC method is simple, rapid, accurate, and sensitive for quantifying eugenol, linalool, and eucalyptol in laurel oil with good repeatability and recovery, which can provide a scientific basis for the quality control and evaluation of laurel oil.
