摘 要:目的考察鹿角方中补骨脂素、异补骨脂素、马钱苷、柚皮苷、橙皮苷、淫羊藿苷、朝藿定A、朝藿定B、朝藿定C、宝藿苷Ⅰ在慢性心力衰竭大鼠左右肾脏中的组织分布。方法经腹主动脉缩窄建立慢性心力衰竭模型后,大鼠灌胃给予鹿角方混悬液(0.16、0.31、0.63 g/mL)30周。LC-MS/MS法测定各成分含有量,分析采用Phenomenex Luna-C18色谱柱(2.0 mm×100 mm,5μm);流动相水-乙腈-0.1%甲酸,梯度洗脱;体积流量0.8 mL/min;柱温40℃;电喷雾离子源正离子扫描模式,多反应监测。结果10种成分在各自范围内线性关系良好(r>0.9990),相对回收率86.53%~111.96%,基质效应RSD 1.09%~12.66%。给药2 h后,同一剂量组大鼠左右肾脏中各成分含有量无明显差异(P>0.05),但除了宝藿苷Ⅰ外,左肾中其他9种成分平均含有量低于右肾中。各成分含有量随着给药剂量增加而升高,线性拟合度大于0.8825。结论该方法稳定可靠,可用于研究鹿角方治疗慢性心力衰竭的药效物质基础。AIM To investigate the tissue distribution of psoralen,isopsoralen,loganin,naringin,hesperidin,icariin,epimedin A,epimedin B,epimedin C and baohuoside I of Lujiao Decoction in left and right kidneys of rats with chronic heart failure.METHODS The models for chronic heart failure were established by abdominal aorta banding,after which rats were given intragastric administration of Lujiao Decoction suspensions(0.16,0.31,0.63 g/mL)for 30 weeks.LC-MS/MS was adopted in the content determination of various constituents.The analysis was performed on a 40℃thermostatic Phenomenex Luna-C18 column(2.0 mm×100 mm,5μm),with the mobile phase comprising of water-acetonitrile-0.1%formic acid flowing at 0.8 mL/min in a gradient elution manner,and electrospray ionization source was scanned in positive ion mode with multiple reaction monitoring.RESULTS Ten constituents showed good linear relationships within their own ranges(r>0.9990),whose relative recoveries were 86.53%-111.96%with the matrix effect RSDs of 1.09%-12.66%.At 2 h after administration,the contents of various constituents in left and right kidneys of rats demonstrated no obvious differences in the same dose group(P>0.05),but the average contents of the other nine constituents(except for baohuoside I)in left kidneys were lower than those in the right kidneys.The contents of various constituents were increased with the enhancement of administration dose,whose linear fitting degrees were more than 0.8825.CONCLUSION This stable and reliable method can be applied to studying the pharmacodynamic substance basis of Lujiao Decoction in treating chronic heart failure.
