摘 要:为了研究不同工艺来源的牛至提取物对大肠杆菌所致小鼠腹泻的防治作用,试验将Balb/c小鼠随机分成7组,分别为正常对照组、模型对照组、枫蓼肠胃康对照组、牛至水提物组、牛至挥发油组、牛至水油组合物组和牛至醇提物组。正常对照组和模型对照组小鼠灌胃生理盐水,枫蓼肠胃康对照组小鼠灌胃给予枫蓼肠胃康颗粒,牛至水提物组、牛至挥发油组、牛至水油组合物组和牛至醇提物组小鼠分别经口给予牛至水提物、牛至挥发油、牛至水油组合物、牛至醇提物,1 d 1次,连用5 d。末次给药30 min后,正常对照组小鼠腹腔注射生理盐水,其余各组小鼠腹腔注射大肠杆菌O101菌液(浓度为4.0×108 cfu/mL),注射量为按体重0.2 mL/10 g。观察小鼠精神状态、腹泻情况及十二指肠组织形态学变化,计算稀便率、稀便级、腹泻指数和腹泻率,评价不同工艺制备的牛至提取物对小鼠腹泻的防治效果。结果表明:枫蓼肠胃康对照组和牛至不同提取物组小鼠精神状态明显优于模型对照组,并且表现出不同程度的腹泻时间延后情况,其中牛至挥发油组、水油组合物组小鼠精神状态最佳。牛至挥发油组、牛至水油组合物组小鼠腹泻率仅为40%,稀便率、平均稀便级和腹泻指数均低于牛至水提物组、牛至醇提物组及枫蓼肠胃康对照组,且以牛至水油组合物的药效最佳。枫蓼肠胃康颗粒和不同工艺来源的牛至提取物都能在一定程度上降低大肠杆菌所致十二指肠肠道损伤,改善小鼠肠道组织结构,其中以牛至水油组合物组小鼠肠组织损伤最小、结构最完整。说明牛至水油组合物对大肠杆菌所致小鼠腹泻防治效果最优。To study the preventive effects of Origanum vulgare L. extracts from different processes on diarrhea in mice infected with E. coli. Balb/c mice were randomly divided into seven groups. Except for model control mice, other groups of mice were orally administered with Fengliao changweikang granules, water extract, volatile oil, alcohol extract and water-oil mixture of Origano once a day for 5 days, respectively. Thirty minutes after the last administration, mice were intraperitoneally injected with E.coli O101 strain(4.0×108 cfu/mL). The injection volume was 0.2 mL/10 g. A normal control group without medicine administration and E.coli injection was also used in the experiments. The mental state, diarrhea related indicators(loose stool rate, loose stool grade, diarrhea index, diarrhea rate) and the changes of duodenal histomorphology were observed. Evaluate the control effect of Oregano extract prepared by different techniques on diarrhea in mice. The results showed that the mental state of the mice treated with Fengliao changweikang and the different extracts of Oregano was significantly better than that of model control group, and the time of diarrhea was delayed to different degrees. The Oregano volatile oil and water-oil mixture group mice had the best mental state. The diarrhea rate of mice treated with Oregano volatile oil and water-oil mixture was only 40%, and their loose stool rate, loose stool grade and diarrhea index were all lower than those in mice treated with Oregano water extract, alcohol extract and Fengliao changweikang group. the preventive effect of Oregano water-oil mixture was the best. Both Fengliao changweikang granules and the extracts of Oregano from different processes could reduce the duodenal intestinal damage induced by E. coli to a certain extent, and repair and improve mouse intestinal tissue structure.
