摘 要:目的探究过氧化物酶5(PRX5)过表达对博来霉素诱导的肺纤维化大鼠的作用及机制。方法50只SD大鼠随机分为五组:对照组、模型组、药物组、空载体组和过表达组,每组各10只。采用博来霉素气管内滴注法构建肺纤维化大鼠模型。HE和Masson染色观察肺组织病理变化;实时荧光定量PCR法检测大鼠肺组织中PRX5 mRNA表达量;酶联免疫吸附法检测肺组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)及羟脯氨酸(HYP)水平;蛋白质免疫印迹法检测肺组织中PRX5、转化生长因子-β1(TGF-β1)、信号转导与转录活化因子3(Stat3)蛋白水平。结果与对照组比较,模型组大鼠肺组织中PRX5 mRNA、SOD、GSH和PRX5水平降低,MDA、HYP、TGF-β1和Stat3水平升高,差异均有统计学意义(均P<0.05)。与模型组比较,过表达组大鼠肺组织中PRX5mRNA、SOD、GSH和PRX5水平升高,MDA、HYP、TGF-β1和Stat3水平降低,差异均有统计学意义(均P<0.05)。结论PRX5过表达对博来霉素诱导的肺纤维化大鼠有一定保护作用,其机制可能与调控TGF-β1和Stat3表达有关。Objective To investigate the effect and mechanism of peroxidase 5(PRX5)overexpression on bleomycin-induced pulmonary fibrosis in rats.Methods Fifty SD rats were randomly divided into five groups:control group,model group,drug group,empty vector group and overexpression group,with 10 rats in each group.A rat model of pulmonary fibrosis was established by intratracheal instillation of bleomycin.HE and Masson staining were used to observe the pathological changes of lung tissue.Real-time fluorescence quantitative PCR was used to detect the expression of PRX5 mRNA in rat lung tissue.Enzyme-linked immunosorbent assay was used to detect malondialdehyde(MDA)and superoxide dismutase(SOD),glutathione(GSH)and hydroxyproline(HYP)levels.Western blot to detect PRX5,transforming growth factor-β1(TGF-β1),signal transducers and activators of transcription 3(Stat3)levels in lung tissue.Results Compared with the controlgroup,the levels of PRX5 mRNA,SOD,GSH,and PRX5 in the lung tissue of the model group decreased,and the levels of MDA,HYP,TGF-β1,and Stat3 increased,and the differences were statistically significant(P<0.05).Compared with the model group,the levels of PRX5 mRNA,SOD,GSH,and PRX5 in the lung tissue of the overexpression group were increased,and the levels of MDA,HYP,TGF-β1,and Stat3 were reduced,and the difference between the two groups was statistically significant(P<0.05).Conclusion PRX5 overexpression has a protective effect on bleomycin-induced pulmonary fibrosis in rats.The mechanism may be related to the regulation of TGF-β1 and Stat3 expression.
