摘　　要：以宽叶独行菜和钝叶独行菜为材料,采用CTAB法提取总DNA,参照文献中引物对两物种nrITS基因、trnL-trnF基因及psbA-trnH基因进行PCR扩增和测序,获得两物种的3个基因片段,并分析其序列特征。结果显示,nrITS基因序列中A+T含量低于G+C含量,而两叶绿体基因序列的碱基组成则相反,A+T含量显著高于G+C含量。用DNAMAN软件对两独行菜物种进行同源性分析,结果表明宽叶独行菜和钝叶独行菜nrITS、trnL-trnF及psbA-trnH基因序列同源性分别为95.65%、97.97%及99.25%。本研究首次扩增获得宽叶独行菜和钝叶独行菜psbA-trnH基因序列,为独行菜属植物分子遗传学积累了研究资料。Lea ves of Lepidium latifolium and Lepidium obtusum were used as materials,from which the total DNA was extracted by the CTAB method.Primers from references were used to PCR experiments and sequencing of nrITS gene,trnL-trnF gene and psbA-trnH gene of both species.Three gene fragments from both species were obtained and characteristics of these genes were analyzed.The results showed that the A+T content of nrITS sequences was lower than that of G+C,while the A+T content was significantly higher than the G+C content in both two chloroplast sequences.DNAMAN software was used to analyze the homology of the two species based on these three genes,the results indicated that the homology of L.latifolium and L.obtusum based on nrITS gene,trnL-trnF gene and psbA-trnH gene were 95.65%,97.97%and 99.25%,respectively.In this study,the psbA-trnH gene sequences of both species was amplified and obtained for the first time,research data was accumulated for further molecular genetic study of Lepidium plants.
