摘 要:tms5是从温敏核不育系安农S-1中鉴定到的不育基因.为验证tms5也是控制不育系HD9802S育性的关键基因,将R287中野生型TMS5基因通过农杆菌介导的转基因方法导入HD9802S中,通过转基因T0代植株育性观察及T1代单株的共分离检测,互补实验从遗传上证明不育系HD9802S不育基因即是tms5.同时本研究通过基因编辑的方法敲除R287中TMS5基因,转基因T0代单株在高温下表现为不育,说明选择合适的受体材料,通过基因编辑技术敲除TMS5基因可以创建新的不育系.tms5 identified from the thermo-sensitive genic male sterile(TGMS)line AnnongS-1 cause the TGMS trait.In order to verify that the TGMS trait of the sterile line HD9802S also indeed corresponds to tms5,genomic DNA fragment harbouring wild-type TMS5 from R287 was transformed into HD9802S by Agrobacterium mediated transgenic method.After checking the fertility of the T0 generation transgenic lines and analysing the co-segregation of the T1 generation transgenic lines,we considered tms5 to cause the sterility at the restrictive temperature through the genetic complementary assay in this study.Moreover,we modified the gene TMS5 of R287 by employing the CRISPR/Cas9 gene editing technology and the trangenic T0 generation lines showed fully sterile under the restrictive condition,this result indicated that we could create new TGMS lines by targeted mutagenesis of TMS5 gene from suitable receptor materials using gene editing technology.
