摘 要:本研究合成三苯胺-卟啉锌复合物(ZnTPor)和PEG-四苯卟啉化合物(PPor),并利用紫外-可见光吸收光谱、荧光发射光谱研究他们与DNA的结合模式.发现两种卟啉化合物均能与DNA有较好结合,PPor为外部结合模式,ZnTPor为嵌插结合模式.ZnTPor、PPor与ct-DNA的结合常数分别为2.17×106和2.63×105(L/mol).也进行了两种化合物的单线态氧测试,结果表明,单线态氧产生效率ZnTPor>PPor.这些研究为后续光动力、光热治疗研究奠定一定的理论基础.Triphenylamine-porphyrin zinc complex(ZnTPor)and PEG-tetraphenylporphyrin compound(PPor)were synthesized,and their binding modes with DNA were studied by UV-vis and fluorescence emission spectra.It was found that both porphyrins could bind to DNA well,PPor was the external binding mode,ZnTPor was the intercalation binding mode.The binding constants of ZnTPor,PPor and ct-DNA were 2.17×106 and 2.63×105(L/mol),respectively.The singlet oxygen test of the two compounds was also carried out.The results showed that the singlet oxygen production efficiency was ZnTPor>PPor.These studies lay a theoretical foundation for the follow-up study of photodynamic and photothermal therapy.
