摘 要:以独叶草天然群体为试材,采用SCoT分子标记技术,研究独叶草天然群体种质资源的遗传多样性,旨在为独叶草种质资源的评价和保护提供分子生物学参考依据。结果表明:从30条引物中筛选出20条重复性好、条带清晰度高、丰富性好的引物进行PCR扩增;20条引物共扩增出99条DNA谱带,其中多态性谱带为72条,多态性比率为72.73%。36份材料的观察等位基因数介于1.3333~2.0000,平均为1.7245;有效等位基因数介于1.2198~1.8063,平均为1.5331;Nei′s基因多样性介于0.1325~0.4390,平均为0.2977;Shannon′s信息指数介于0.1956~0.6289,平均为0.4324。聚类分析表明,36份独叶草种质资源的遗传相似系数在0.6500~0.9490,平均遗传相似性系数为0.7577;而编号相邻或相近的独叶草种质资源均聚类在一个大类或亚类。这说明SCoT标记可适用于独叶草的遗传多样性分析。The natural population Kingdonia uniflora Balf.F.et W.W.Smith were used as test materials,by using SCoT markers,was studied to estimate genetic diversity of the natural population Kingdonia uniflora Balf.F.et W.W.Smith,aiming at providing molecular biological basis for the evaluation and protection of the germplasm resources of Kingdonia uniflora Balf.F.et W.W.Smith.The results indicated that selected 20 reproducible and clear bands from 30 primers were amplified by PCR,a total of 99 bands were amplified by 20 primers,including 72 polymorphic bands,and the polymorphic rate was 72.73%.The observed number of alleles in 36 materials was from 1.333 3 to 2.000 0,with an average of 1.724 5.The effective number of alleles was from 1.219 8 to 1.806 3,with an average of 1.533 1;Nei′s gene diversity was from 0.132 5 to 0.439 0,with an average of 0.297 7.Shannon information index was from 0.195 6 to 0.628 9,with an average of 0.432 4.The cluster analysis showed that the genetic similarity coefficient of 36 Kingdonia uniflora Balf.F.et W.W.Smith germplasm resources was from 0.650 0 to 0.949 0,with an average of 0.757 7.The Kingdonia uniflora Balf.F.et W.W.Smith germplasm resources of the neighbouring or similar identifier were clustered in a categorie or subclass.This indicated that genetic diversity of Kingdonia uniflora Balf.F.et W.W.Smith germplasm could be revealed by SCoT markers.
