摘 要:以剑河双钩藤为试材,比较了微波辅助萃取、超声波辅助萃取、超声-微波协同萃取3种方法对钩藤碱得率的影响,在单因素试验基础上,采用Box-Behnken响应面法优化超声-微波协同萃取剑河双钩藤钩藤碱提取工艺,以期得到超声-微波协同萃取钩藤碱的最佳工艺参数。结果表明:超声-微波协同萃取法的钩藤碱得率远高于超声波、微波萃取法。最佳超声-微波协同萃取工艺参数为萃取时间15 min、乙醇体积分数46%、料液比1∶208 g·mL-1,超声波功率150 W,微波功率150 W。在该条件下,剑河双钩藤钩藤碱的得率为52.9281 mg·g-1,与模型预测值52.9579 mg·g-1相近。优选的超声-微波协同萃取工艺提高了剑河双钩藤中钩藤碱得率。Rhynchophylline from Uncaria laevigata Wal.ex G.Don as test material,the effects of microwave-assisted extraction,ultrasound-assisted extraction and ultrasound-microwave synergistic extraction on the yield of rhynchophylline were compared,on the basis of single factor experiment,the optimum ultrasound-microwave synergistic extraction process of the rhynchophylline from Uncaria laevigata Wal.ex G.Don in Jianhe was analyzed by using Box-Behnken design(BBD) with response surface methodology,in order to obtain the optimal ultrasonic-microwave synergistic extractionparameters for rhynchophylline.The results showed that the yield of rhynchophylline by ultrasonicmicrowave synergistic extraction was much higher than that of ultrasonic-assisted and microwaveassisted extraction.The optimal ultrasonic-microwave synergistic extraction parameters for rhynchophylline from Uncaria laevigata Wal.ex G.Don in Jianhe were determined as follows15 minutes of extraction time,46% of ethanol volume fraction,1∶208 g·mL-1 of solid-liquid ratio,150 W of ultrasonic powder,150 W of microwave powder.Under these conditions,the yield of rhynchophylline fromUncaria laevigata Wal.ex G.Don in Jianhe was 52.928 1 mg·g-1,which was cloded to the model predicted value of 52.957 9 mg·g-1.The optimal ultrasound-microwave synergistic extraction process improved the yield of rhynchophylline fromUncaria laevigata Wal.ex G.Don in Jianhe.
