摘  要:以葡萄糖为对照品,利用苯酚-硫酸显色法,采用紫外-可见分光光度法测定藏药唐古特青兰多糖含量,用单因素试验法与响应面法确定溶剂萃取-超声波辅助提取多糖的优化工艺。结果显示:在溶剂萃取仪压力为300 psi、超声波功率为200 W、提取温度为52℃、提取时间为80 min、液料比为8 mL/mg的条件下,多糖的平均提取率为67.28%,与理论预测值67.36%的相对标准偏差为0.0595%。结论:该优化工艺能显著提高多糖的提取率提取时间短、操作简便、稳定。with glucose as the control,the content of Dracocephalum Tanguticum polysaccharide in Tibetan medicine was determined by phenol sulfuric acid colorimetry and UV-vis spectrophotometry,and the optimization process of solvent extraction ultrasonic assisted extraction polysaccharide was determined by single factor test and response surface methodology.The results showed that when the pressure of solvent extractor was 300 psi,ultrasonic power was 200 W,extraction temperature was 52℃,extraction time was 80 min,and the ratio of liquid and material was 8 mL/mg,the average extraction rate of polysaccharide was 67.28%,and the relative standard deviation with the theoretical prediction value of 67.36%was 0.0595%.Conclusion:The optimized process can significantly improve the extraction rate of polysaccharide,the extraction time is short,the operation is simple and stable.
