摘  要:实验制备5-氟尿嘧啶/蒙脱石缓释药物(5-Fu/Mt),研究5-Fu/Mt缓释药物与蛋白质的相互作用及其对体外培养肺癌A549细胞的影响。研究表明:牛血清白蛋白(BSA)在5-Fu/Mt上的吸附等温曲线呈S型,且10 h后达到吸附/脱附平衡;随着BSA浓度的增加,缓释药物中5-Fu的释放量减小;5-Fu/Mt对体外培养肺癌A549细胞的半数抑制浓度为27.67 ug/mL,高浓度BSA的存在会减弱5-Fu/Mt对肺癌A549细胞生长的抑制作用。The 5-fluorouracil/montmorillonite sustained-release drugs(5-Fu/Mt)were prepared.Besides,the interaction between protein and 5-Fu/Mt and the resulted effect on A549 cells cultured in vitro were analyzed.The results showed that the adsorption isotherm of bovine serum albumin(BSA)on 5-Fu/Mt showed an S-shaped isotherm,and the adsorption/desorption equilibrium was reached after 10 h.The cumulative drug release of 5-Fu from 5-Fu/Mt decreased with increasing BSA concentration.In vitro cell culture experiment results revealed that the half inhibitory concentration of 5-Fu/Mt on lung cancer A549 cells was 27.67μg/mL,and the presence of high concentration of BSA weakened the growth inhibitory effect of 5-Fu/Mt on A549 cells.
