摘 要:目的:探讨蛴螬提取物对兔光损伤视网膜变性Caspase-8、Bax、Bid、Caspase-3 mRNA表达的影响。方法:将30只雄性实验性兔随机平均分为空白组、模型组、蛴螬低剂量组、蛴螬中剂量组和蛴螬高剂量组。除空白组外,其余4组均建立光损伤视网膜变性模型,在给药14 d处死后取材采用RT-qPCR法测定Caspase-8 mRNA、Bax mRNA、Bid mRNA、Caspase-3 mRNA的表达水平。结果:与模型组比较,蛴螬低、中、高剂量组的Caspase-8 mRNA、Bax mRNA、Bid mRNA、Caspase-3 mRNA相对表达量降低,差异具有统计学意义(P<0.05);与蛴螬低剂量组比较,中、高剂量组的Bax mRNA和Caspase-3 mRNA改善更为明显(P<0.05),但中、高剂量组之间没有统计学意义(P>0.05)。结论:蛴螬通过抑制Caspase-8 mRNA、Bax mRNA、Bid mRNA、Caspase-3 mRNA的表达,抑制细胞凋亡受体信号通路来发挥保护视网膜作用,中、高剂量组效果较好。Objective:To investigate the effects of grub extract on the mRNA expressions of Caspase-8,Bax,Bid and Caspase-3 in rabbits with optical damage and retinal degeneration.Methods:30 experimental rabbits were randomly divided into the blank group,the model group,the grub groups of low-dose,medium-dose and high-dose,with 6 rabbits(12 eyes)in each group.The model of optical damage and retinal degeneration was established.The mRNA expressions of Caspase-8,Bax,Bid and Caspase-3 were determined by RT-qPCR after 14 days of intervention.Results:Compared to the model group,the relative mRNA expressions of Caspase-8,Bax,Bid and Caspase-3 were significantly increased in the grub groups of low-dose,medium-dose and high-dose(P<0.05).Compared to low-dose group,the improvements of Bax mRNA and Caspase-3 mRNA were more significant in the medium-group and high-dose group(P<0.05);of which there were no statistical differences between the medium-dose group and the high-dose group(P>0.05).Conclusion:Grub extraction can inhibit the mRNA expressions of Caspase-8,Bax,Bid and Caspase-3 and inhibit the apoptotic receptor signaling pathway to protect retina.The groups of medium-dose and high-dose have better effects.
