摘  要:目的:观察自拟醒神汤对血管性痴呆大鼠炎性细胞因子TNF-α、IL-1β表达水平的影响。探讨血管性痴呆大鼠炎症损伤机制及自拟醒神汤对血管性痴呆大鼠脑保护作用机制。方法:采用自体血栓法制作血管性痴呆模型。随机分为正常组、模型组、中药组和西药组。Morris水迷宫实验检测大鼠的空间学习以及记忆能力;ELISA法检测大鼠海马区TNF-α、IL-1β表达水平。结果:与模型组比较,治疗组的行为学症状较模型组明显改善。与模型组比较,治疗组海马区TNF-α、IL-1β表达水平明显下降。结论:促炎因子TNF-α、IL-β在缺血性脑损伤中起到重要作用。自拟醒神汤能够抑制炎性细胞因子释放,减轻脑组织损伤,改善行为学症状。Objective:To observe the effect of self-made Xingshen Decoction on the expressions of TNF-αand IL-1βin rats with vascular dementia(VD),so as to study the mechanism of inflammation damage in VD rats and the protective effect of self-made Xingshen Decoction on the brain of VD rats.Methods:The rats were randomly divided into the normal group,the model group,the Chinese medication group and the western medication group.The VD model was made by autologous thrombus.Morris water maze test was used to detect spatial learning and memory ability of the rats,and ELISA method was applied to detect the expressions of TNF-αand IL-1βin hippocampus of the rats.Results:Compared to the model group,the behavioral symptoms improved significantly and the expression levels of TNF-αand IL-1βin the hippocampus decreased significantly in the Chinese medication group.Conclusion:Inflammatory cytokines of TNF-αand IL-1βplay important roles in ischemic brain damage.Self-made Xingshen Decoction can inhibit the release of inflammatory cytokines,alleviate brain tissue damage and improve behavioral symptoms.
