摘  要:目的:通过对术后肠麻痹小鼠的动物实验和转录组学研究,阐明电针干预术后肠麻痹的疗效机制。方法:选取小鼠术后肠麻痹模型为研究对象,随机分为假手术组、模型组和电针组,于电针处理后24 h取小肠组织,通过转录组测序技术筛选差异表达基因,并对差异基因进行GO功能注释和KEGG通路富集分析。结果:在生物信息学分析方面,模型组与假手术组比较,上调差异表达基因161个,下调差异表达基因327个;电针组与模型组比较,上调差异表达基因25个,下调差异表达基因242个。结合GO和KEGG分析结果,筛选得到可能与电针治疗术后肠麻痹小鼠相关的3个信号通路:细胞因子-细胞因子-受体互作通路、肿瘤坏死因子信号通路和神经活性配体与受体互作通路。结论:电针可能是通过改变细胞趋化性、白细胞迁移、炎症反应、趋化因子受体结合和趋化因子活性改变了术后肠麻痹小鼠肠内的炎症水平和微环境,从而恢复肠道转运功能。Objective:To elucidate the therapeutic mechanism of electro-acupuncture intervention for postoperative ileus in mice through animal experiments and transcriptomic studies.Methods:The mice were randomly divided into the sham operation group,the model group and the electro-acupuncture group.The model of postoperative ileus was established in mice.The intestine tissues were collected 24 h after electro-acupunctre intervention,differentially expressed genes were screened out by transcriptome sequencing technique,GO funcitional annotation and the enrichment analysis of KEGG pathyway were carried out afterwards.Results:In terms of bioinformatics analysis,161 differentially expressed genes were up-regulated and 327 differentially expressed genes were down-regulated in the model group compared to those in the sham operation group;25 differentially expressed genes were up-regulated and 242 differentially expressed genes were down-regulated in the electro-acupuncture group compared to those in the model group.Combined with the results of GO and KEGG analysis,three signaling pathways that might be associated with electro-acupuncture treatment in postoperative ileus mice were screened,they were cytokine-cytokine-receptor interaction pathway,tumor necrosis factor signaling pathway and neuroactive ligand and receptor interaction pathway.Conclusion:Electro-acupuncture may restore intestinal transport function by altering cell chemotaxis,leukocyte migration,inflammatory response,chemokine receptor binding and chemokine activity to change the level of inflammation and microenvironment in the intestines of postoperative ileus mice.
