摘 要:[目的]本文旨在评价山西本土草地早熟禾(Poa pratensis L.)种质资源的遗传多样性和遗传结构。[方法]本研究利用10对ISSR引物对采集于10个野生居群的49份种质资源样品和1份商品种‘爱美(Aimyth)’进行遗传差异分析,利用PopGen1.32软件计算各居群间及样品间的遗传相似系数、基因多样性指数、香农信息指数等遗传参数,利用GenAlEx 6.5软件计算居群间多态位点比例,利用NTSYSpc-2.10e软件构建亲缘关系聚类图。[结果]10条ISSR引物共扩增出250条谱带,引物多态性比率为94.80%。11个居群间的遗传相似系数为0.5800~0.9467。据UPGMA聚类分析,在遗传相似系数0.75处,11个居群共分为5类。其中与大部分供试样品地理位置较远的右玉居群、屯留居群和商品种‘爱美’为单独一类。在11个居群中,其中样品数较丰富的7个居群基因多样性指数介于0.0971~0.2470之间,而香农信息指数介于0.1398~0.3683之间,多态位点比例介于22.80%~70.00%之间。此7个居群内的遗传多样性水平差异较大,其中芽庐山居群内的遗传多样性水平最高,五台山居群内的遗传多样性水平最低。[结论]山西地区草地早熟禾具较高的遗传多样性,不同居群间的遗传多样性水平存在差异,野生居群与商品种‘爱美’间均存在较大的遗传距离。[Objective]This study aimed to evaluate the genetic diversity and genetic structure of the germplasm resources of Poa pratensis L.in Shanxi.[Methods]A total of 49 wild samples were collected and divided into ten populations.Inter-simple Sequence Repeat(ISSR)was used and a commercial cultivar‘Aimyth’was selected as a referenced outgroup.The genetic differences of 50 tested samples were analyzed by 10 pairs of ISSR polymorphic primers.The statistical analyses were conducted by software GenAlEx 6.5,Pop Gen 1.32 and NTSYSpc-2.10 e.[Results]A total of 250 bands were amplified by 10 ISSR primers,and the primer polymorphic ratio was 94.8%.The genetic similarity coefficients among these 11 populations was 0.5800~0.9467.According to UPGMA cluster analysis,the 11 populations were divided into 5 categories based on the genetic similarity coefficient at the level of 0.75.In the cluster analysis,the groups of YY and TL,which were geographically far away from most of the tested samples,and commercial species‘Aimyth’were separated into one group.Among the 11 population groups,the genetic diversity index of seven populations with the richer number of samples ranged from 0.0971 to 0.2470,and the Shannon’s information index ranged from 0.1398 to 0.3683.The percentage of polymorphic loci was between22.80%and 70.00%.The genetic diversity levels of the seven population groups were quite different,Among them,the group YL had the highest genetic diversity level and the group WT was the lowest one.[Conclusion]The genetic diversity of Poa pratensis L.in Shanxi was abundant.There were different genetic diversity levels among different populations,and existed a large genetic distance between wild population and‘Aimyth’.
