摘  要:目的了解1株人流感H3N2毒株的全基因组序列特征、遗传进化及生物学特性。方法对浙江省丽水市分离的1株H3N2流感毒株进行全基因组测序,从美国国立生物信息中心(NCBI)和全球倡议分享流感数据库(GISAD)获取其他流感毒株基因组全序列,通过BioEdit7.0.9将该毒株8节段基因与其他序列比对,MEGA7.0软件邻接法构建进化树,并全面分析该毒株的进化、致病力和抗原性等特征。结果测序得到该毒株为人流感分离株A/Homo sapien/China/LS340/2019(H3N2)株(LS340),LS340为典型的季节性人流感病毒,血凝素基因属于3C.2a1进化分支,具备人际传播能力,部分位点呈现高传播性和高致病性特征,对金刚烷胺耐药。与当季推荐疫苗株相比,抗原区出现涉及A、B两个表位3个位点突变。结论LS340未出现重组、缺失等较大变异,但与当季推荐疫苗株相比已经发生抗原漂变,应及时更新疫苗,同时加强此类病毒变异监测,防止流感大流行发生。Objective To study the whole genome sequence,genetic evolution status and biological characteristics of an H3N2 subtype influenza virus.Methods The whole genome sequence of a strain of H3N2 subtype influenza virus isolated in Lishui,Zhejiang was obtained.The other genome sequences of influenza strains were acquired from National Center for Biotechnology Information(NCBI)and Global Initiative on Sharing All Influenza Date(GISAID).The eight gene segments of the strain were compared with the referenced virus sequence by BioEdit7.0.9 software.The phylogenetic evolution tree was constructed by Neighbor-Joining method with Mega5.0 software,then the characteristics of virus evolution,pathogenicity and infectivity were analyzed.Results The sequence analysis showed the strain was human influenza A virus A/Homo sapien/China/LS340/2019(LS340).LS340 was a typical seasonal influenza virus.It belonged to the 3C.2a1 branch of genetic evolution according to hemagglutinin gene,and had the transmission ability among individuals.Some sites had the characteristics of high transmission and high pathogenicity,and the strain was resistant to amantadine.Compared with the vaccine strain,LS340 had 3 amino acid mutations involved in A and B antigen regions.Conclusions LS340 has no major variation,such as recombination or deletion,but multiple mutations occur in the key domains of amino acids leading to antigenic drift.The vaccine strain should be updated in time,meanwhile the surveillance of such virus variances should be strengthen to prevent an outbreak of influenza pandemic.
