摘 要:目的:探讨电针“委中”对多裂肌损伤模型大鼠CaM/CaMKII/iNOS信号通路介导的海马氧化应激的影响。方法:SD大鼠随机选取8只作为空白对照组,剩余大鼠采用L4~5多裂肌注射布比卡因造模后随机分为模型1 d组、模型3 d组、电针1 d组及电针3 d组,每组各8只。空白组和模型组不做任何治疗。电针组大鼠双侧“委中”电针,每次20 min,每日1次。分别于治疗的第1天和第3天同步取材。观察各组大鼠海马组织中CaM、CaMKII、iNOS的含量和蛋白表达,MDA的浓度和SOD的活性。结果:与空白组比较,模型1 d组和模型3 d组海马组织中CaM、CaMKII、iNOS的含量和表达升高(P<0.01或P<0.05),MDA浓度升高、SOD活性降低(P<0.01);与模型1 d组比较,电针1 d组海马组织中CaM、CaMKII、iNOS的含量、CaM、iNOS的蛋白表达量变仅明显(P<0.05或P<0.01),MDA浓度降低、SOD活性升高(P<0.01);与模型3 d组比较,电针3 d组海马组织中CaM、CaMKII、iNOS含量、CaM、CaMKII蛋白表达量变化明显(P<0.01或P<0.05),MDA的浓度降低(P<0.01);与电针1 d组比较,电针3 d组海马组织中CaM、CaMKII含量、iNOS蛋白表达量降低(P<0.01或P<0.05),CaM、CaMKII的蛋白表达量升高(P<0.05),iNOS含量、MDA浓度和SOD活性差异无统计学意义(P>0.05)。结论:电针“委中”可能通过抑制CaM/CaMKII/iNOS通路提高抗氧化能力减轻多裂肌损伤大鼠模型海马组织的氧化应激损伤。Objective:To study the effect of electro-needling Weizhong(BL40)on oxidative stress in hippocampus mediated by CaM/CaMKII/iNOS signaling pathway in rats with multifidus injury.Methods:40 SD rats were randomly into the blank group,the model 1D group,the model 3D group,the EA 1D group and the EA 3D group,with 8 rats in each group.The model was established by injection of bupivacaine into L4-L5 multifidus.The blank group and the model group did not receive any treatments.The rats in the EA groups were intervened by electro-needling BL40,20 minutes per day.Samples were collected on the first and third day of treatment.The contents and expressions of CaM,CaMKII and iNOS,the concentration of MDA and the activity of SOD in hippocampus were observed in each group.Results:Compared to the blank group,the content and expression of CaM,CaMKII and iNOS in hippocampus significantly increased in the model 1D group and the model 3D group(P<0.01,P<0.05);the concentration of MDA increased and the activity of SOD decreased(P<0.01).Compared to the model 1D group,the contents of CaM,CaMKII and iNOS,the protein expression of CaM and iNOS in hippocampus decreased in the EA 1D group(P<0.05 or P<0.01),the concentration of MDA decreased,and the activity of SOD increased(P<0.01).Compared to the model 3D group,the contents of CaM,CaMKII,iNOS,and the proteins expressions of CaM and CaMKII in hippocampus decreased in the EA 3D group(P<0.01 or P<0.05);the concentration of MDA decreased(P<0.01).Compared to the EA 1D group,the contents of CaM and CaMKII,as well as the protein expression of iNOS in hippocampus decreased in the EA 3D group(P<0.01 or P<0.05);the protein expressions of CaM and CaMKII increased(P<0.05);there were no significant differences in iNOS content,MDA concentration and SOD activity between the EA 1D group and the EA 3D group(P>0.05).Conclusion:Electro-needling BL40 may alleviate hippocampus oxidative stress injury in rats with multifidus injury by inhibiting CaM/CaMKII/iNOS pathway and improving antioxidant capacity.
