摘 要:目的:在体外实验条件下,评价聚磷酸钙(CPP)-淫羊藿苷(ICA)复合骨支架的细胞生物相容性。方法:将CPP-ICA按质量4∶1制作出复合骨支架,按比例1 g复合骨支架浸提介质10 ml,浸提介质为包含体积分数为10%胎牛血清的DMEM,制备出骨支架的浸提液,并用MG63细胞与复合骨支架及浸提液共同培养。在不同时间点,用倒置显微镜观察细胞形态学改变,细胞生长增殖情况用MTT法检测。结果:测定时间点细胞在CPP/ICA复合骨支架表面生长良好,增殖迅速,细胞形态正常。MTT法检测显示,随着培养时间的延长,实验组和阴性对照组细胞光吸收值(A值)逐渐增高,且增高显著(P=0.000),两组之间A值差异无统计学意义(P>0.05);培养24 h,实验组和阴性对照组的A值高于阳性对照组,但差异无统计学意义(P>0.05),培养48 h、72 h,实验组和阴性对照组A值明显高于阳性对照组(P=0.000),细胞毒性为0级。结论:CPP-ICA复合骨支架具有良好的细胞生物相容性,无细胞毒性,可用于动物体内试验。Objective To observe the biocompatibility of CPP-ICA composited bone scaffold by the experiment in vitro.Method The composited bone scaffold was made by CPP-ICA at the mass of 4∶1.At the proportion of 1 g composited bone scaffold,the extraction medium was 10 ml.the medium was DMEM(Dulbecco's modified Eagle medium)included volume fraction of 10%fetal bovine serum.The extraction solution of the composited bone scaffold was prepared,MG63 cells were co-cultured with the composited bone scaffold and the extraction solution.At different time points,to observe Cell morphological changes by inverted microscope,and cell growth and proliferation by MTT assay.Results At the different time points of the test,the cells grew well on the surface of the CPP/ICA composited bone scaffold,proliferated rapidly,and the cell morphology was normal.With the extension of culture time MTT assay showed that,the light absorption value(A value)of cells in the experimental group and the negative control group increased gradually and significantly(P=0.000),and the difference of A value between the two groups was not significant(P>0.05).after 24 h,the A value of the experimental group and the negative control group was higher than that of the positive control group(P>0.05),after 48 h and 72 h,the A value of the experimental group and the negative control group was significantly higher than that of the positive control group(P=0.000).the cytotoxicity was 0.Conclusion The CPP-ICA composited bone scaffold has good biocompatibility and no cytotoxicity,and can be used to animaltest in vivo.
