摘 要:目的探讨活性维生素D3(Vit D3)对T2DM大鼠肝脏的保护作用及作用机制。方法SD雄性大鼠分为正常对照(NC)组、T2DM组和活性Vit D3干预组(Vit D3组),Vit D3组予骨化三醇灌胃,NC、T2DM组予花生油灌胃,8周后处死各组大鼠,测量血清血糖、TC、TG、谷草转氨酶(AST)、谷丙转氨酶(ALT)水平;HE染色观察肝脏组织的形态学改变;免疫组织化学法、荧光定量RT-PCR检测肝脏核因子κB(NF-κB)、单核细胞趋化蛋白1(MCP-1)、细胞间黏附因子1(ICAM-1)、转化生长因子β1(TGF-β1)的蛋白及mRNA表达水平。结果Vit D3组血清TG、ALT、AST水平低于T2DM组(P<0.05)。Vit D3组肝脏病理组织形态学,尤其是组织结构完整性优于T2DM组。Vit D3组NF-κB、MCP-1、ICAM-1、TGF-β1的蛋白和mRNA表达水平低于T2DM组(P<0.05)。NF-κB的mRNA表达水平与MCP-1、ICAM-1、TGF-β1 mRNA表达水平呈正相关(r=0.711、0.734、0.698,P<0.01)。结论活性Vit D3可能通过NF-κB介导的炎症途径对T2DM大鼠肝脏发挥保护作用。Objective To investigate the protective effects and mechanisms of active vitamin D3(Vit D3)on liver injury in diabetic rats.Methods The SD male rats were divided into normal control group(NC group),diabetes group(T2 DM group)and Vit D3 intervention group(Vit D3 group).The Vit D3 group was fed with calcitriol,and the NC group and T2 DM group were fed with nut oil.Eight weeks later all rats were killed,the levels of serum total cholesterol(TC),triglyceride(TG),alanine aminotransferase(ALT),aspartate aminotransferase(AST)were detected.Morphological changes of liver tissue was tested by HE staining.The protein and mRNA expressions levels of liver core transcription factors(NF-κB),monocyte chemoattractant protein 1(MCP-1),intercellular adhesion molecule 1(ICAM-1),transforming growth factorβ1(TGF-β1)were detected by immunohistochemistry and real-time quantitative reverse transcription polymerase chain reaction assay(RT-PCR).Results The levels of serum TG,ALT and AST were lower in Vit D3 group than in T2 DM group(P<0.05).The pathological morphology,especially the integrity of tissue structure was better in Vit D3 group than in T2 DM group.The protein and mRNA expression levels of NF-κB,MCP-1,ICAM-1 and TGF-β1 were lower in Vit D3 group than in T2 DM group(P<0.05).The mRNA expression level of NF-κB was positively correlated with the mRNA expression levels of MCP-1,ICAM-1 and TGF-β1(r=0.711,0.734,0.698,P<0.01).Conclusion The active Vit D3 may play a protective role in the liver of T2 DM rats by NF-κB-mediated inflammatory pathways.
