摘 要:为明确贵州白山羊ADIPOQ基因及其编码蛋白的结构及功能,本实验通过PCR技术拼接白山羊外显子序列,分析其生物信息学特征及在公母羊不同组织中的表达特点。结果表明:白山羊ADIPOQ基因CDS区长720 bp、编码239个氨基酸,编码蛋白为水溶性蛋白,相对分子量为25 955.14 ku,理论等电点为5.96,且存在17个氨基酸的信号肽区,不属于跨膜蛋白,但属于分泌蛋白,含有16个磷酸化位点,具有1个C1Q保守结构域,主要与ADIPOR1、ADIPOR2、CDH13及LEP蛋白进行信息互作,系统进化分析结果与亲缘关系远近一致。实时荧光定量检测表明,ADIPOQ基因在白山羊各组织中均有表达,其中ADIPOQ基因在肺的表达量最高,其次是脾、心和肝,在肾中表达量较低,且这种表达存在性别差异,ADIPOQ基因在母羊脾脏、肺脏和心脏组织中的表达量较高,而在公羊的肝脏和肾脏中表达量较高。本研究通过对贵州白山羊ADIPOQ基因、编码蛋白的生物信息学及不同组织表达分析,预测了ADIPOQ蛋白分子作用及功能,为进一步分析ADIPOQ基因在白山羊脂肪累积过程中的功能奠定了理论基础。The aim is to study the structure and function of ADIPOQ gene and protein of Guizhou White Goat. The complete sequence of the CDS of ADIPOQ gene was acquired by PCR technique, and to analyze its bioinformatics characteristics and expression characteristics in different tissue of ewe and ram. The results showed that the coding sequence of ADIPOQ gene was 720 bp and encoding 239 amino acids, the encoded protein was water-soluble protein and relative molecular weight was 25 955.14 Da, isoelectric point was 5.96. The protein contained a signal peptide region that consist of 17 amino acids and is non-transmembrane protein. But belongs to the secreted protein and contained 16 phosphorylation sites and 1 conserved domain C1Q. The ADIPOQ protein mainly interacted with ADIPOR1, ADIPOR2, CDH13 and LEP protein. The phyletic evolution was the same as their genetic relationship. The RT-PCR detection showed that ADIPOQ was expressed in all tissues, but the highest was in lung, followed by spleen, heart and liver, and the lowest in kidney, and the expression have difference in ewe and ram, and the expression level was higher in spleen, lung and heart in ewe, but was higher in liver and kidney in ram. Through the bioinformatics and expression analysis in different tissues of the ADIPOQ gene of Guizhou white goat, the molecular action mechanism and function of ADIPOQ protein was predicted, and these studies have laid an information foundation for further study of the ADIPOQ gene function in the goat fat accumulation process.
