摘  要:目的建立HPLC同时测定健儿清解液中绿原酸、咖啡酸、芦丁、金丝桃苷、木犀草苷、陈皮苷、连翘苷、木犀草素、芹菜素的含量。方法采用Dikma C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-体积分数为0.1%磷酸水溶液(B)梯度洗脱,流速:1.0 mL·min^-1,柱温:30℃,波长切换325 nm(0~25 min,测定绿原酸、咖啡酸)、254 nm(25~36 min,测定芦丁、金丝桃苷、木犀草苷)、280 nm(36~50 min,测定陈皮苷、连翘苷)、347 nm(50~60 min,测定木犀草素、芹菜素)。结果绿原酸、咖啡酸、芦丁、金丝桃苷、木犀草苷、陈皮苷、连翘苷、木犀草素、芹菜素9个成分的进样量分别在绿原酸23.09~161.6 mg·L^-1(r=0.9999)、咖啡酸1.400~9.802 mg·L^-1(r=0.9995)、芦丁1.667~11.67 mg·L^-1(r=0.9996)、金丝桃苷2.546~17.82 mg·L^-1(r=0.9998)、木犀草苷4.454~31.18 mg·L^-1(r=0.9999)、陈皮苷8.790~61.53 mg·L^-1(r=0.9997)、连翘苷3.516~24.61 mg·L^-1(r=0.9995)、木犀草素0.3900~2.730 mg·L^-1(r=0.9996)、芹菜素0.4404~3.080 mg·L^-1(r=0.9995)与峰面积呈良好的线性关系;加样回收率(n=3)分别为99.6%(RSD=1.1%)、98.3%(RSD=1.3%)、98.7%(RSD=1.6%)、99.5%(RSD=1.1%)、100.1%(RSD=1.0%)、100.0%(RSD=1.1%)、98.6%(RSD=1.2%)、98.2%(RSD=1.4%)、99.3%(RSD=1.2%)。结论本法可用于健儿清解液的质量控制。Objective To develop an HPLC method for simultaneous determination of chlorogenic acid,caffeic acid,rutin,hyperin,luteoloside,hesperidin,phillyrin,luteolin and apigenin in Jianerqingjieye Oral Liquid.Methods The chromatographic separation was carried out on Dikma C18 column(4.6 mm×250 mm,5μm)with a mobile phase consisting of acetonitrile(A)-0.1%phosphoric acid solution(B)in a gradient mode at a flow rate of 1.0 mL·min^-1 and the column temperature was 30℃.The UV detection wavelength was set at 325 nm for chlorogenic acid and caffeic acid during 0-25 min,254 nm for rutin,hyperin and luteolosid during 25-36 min,280 nm hesperidin and luteolin during 36-50 min,347 nm for luteolin and apigenin during 50-60 min.Results The ranges for linear correlation of chlorogenic acid,caffeic acid,rutin,hyperin,luteolosid,hesperidin,phillyrin,luteolin and apigenin were 23.09-161.6 mg·L^-1(r=0.9999),1.400-9.802 mg·L^-1(r=0.9995),1.667-11.67 mg·L^-1(r=0.9996),2.546-17.82 mg·L^-1(r=0.9998),4.454-31.18 mg·L^-1(r=0.9999),8.790-61.53 mg·L^-1(r=0.9997),3.516-24.61 mg·L^-1(r=0.9995),0.3900-2.730 mg·L^-1(r=0.9996)and 0.4404-3.080 mg·L^-1(r=0.9995),respectively.The average recoveries(n=3)of the nine components were 99.6%(RSD=1.1%),98.3%(RSD=1.3%),98.7%(RSD=1.6%),99.5%(RSD=1.1%),100.1%(RSD=1.0%),100.0%(RSD=1.1%),98.6%(RSD=1.2%),98.2%(RSD=1.4%)and 99.3%(RSD=1.2%),respectively.Conclusion The method can be used for quality control of Jianerqingjieye Oral Liquid.
