摘  要:目的探索中药复方益糖康中黄芪-丹参药对治疗糖尿病肾病的效应机制。方法通过中药系统药理学分析平台(TCMSP)来检索黄芪、丹参两味药物的化学成分和作用的靶点。并在TTD、OMIM、Drugbank、Digsee等多种数据库查询与糖尿病肾病相关的基因。同时基于UniProt数据库分析出靶点多对应的基因,并通过Cytoscape 3.6.0软件构建出黄芪-丹参的化合物-靶点网络、蛋白质相互作用(PPI)网络筛选出核心靶点,最后使用DAVID数据库进行基因本体(GO)功能富集分析和京都基因和基因组百科全书(KEGG)通路富集分析,得出其可能存在的作用机制。结果预测得到黄芪-丹参治疗DN的化合物-靶点网络中有效成分为85个,有效靶点861个,并因此得出其作用机制可能与缺氧诱导因子1(HIF-1)信号通路、表皮生长因子受体(EGFR)酪氨酸激酶抑制剂耐药机制、胰岛素信号通路、MAPK信号通路等有关,并且STAT3、AKT1、PIK3CA、PIK3R1、JAK1、JUN等靶点基因可能起着重要作用。结论通过网络药理学探讨了中药复方益糖康中黄芪-丹参药对治疗糖尿病肾病的多成分-多靶点-多通路的作用机制特点,为进一步揭示中药复方益糖康治疗糖尿病及其并发症的作用机制的科研探索提供了新的思路和新的方法。Objective To explore the effect mechanism of Huangqi(Astragalus);Danshen(Salvia miltiorrhiza)pair in traditional Chinese medicine compound Yitangkang in the treatment of diabetic nephropathy.Methods The Chinese medicine system pharmacology analysis platform(TCMSP)was used to search for the chemical constituents and targets of Huangqi(Astragalus)and Danshen(Salvia miltiorrhiza).And in the TTD,OMIM,Drugbank,Digsee and other databases,we studied the genes associated with diabetic nephropathy.At the same time,based on the UniProt database,the corresponding genes of the target were analyzed,and the compound-target network and protein interaction(PPI)network of Huangqi(Astragalus)-Danshen(Salvia miltiorrhiza)was constructed by Cytoscape 3.6.0 software to screen out the core targets.Finally,the DAVID database was used.Gene ontology(GO)functional enrichment analysis and Kyoto Genomic and Genomic Encyclopedia(KEGG)pathway enrichment analysis were used to study its possible mechanism of action.Results It was predicted that there were 85 active components and 861 effective targets in the compound-target network of Huangqi(Astragalus)-Danshen(Salvia sinensis).Therefore,the mechanism of action may be related to hypoxia-inducible factor 1(HIF-1)signaling pathway.Epidermal growth factor receptor(EGFR)tyrosine kinase inhibitor resistance mechanism,insulin signaling pathway,MAPK signaling pathway and other related genes,and STAT3,AKT1,PIK3 CA,PIK3 R1,JAK1,JUN and other target genes may play an important role.Conclusion The mechanism of multi-component-multi-target-multi-channel treatment of diabetic nephropathy with traditional Chinese medicine compound Yitangkang was explored by network pharmacology,in order to further reveal its concurrent scientific research and provide new ideas and new methods for exploration of the mechanism of the traditional Chinese medicine compound Yitangkang in the treatment of diabetes.
