摘  要:目的分析自发性高血压大鼠(SHR)肠道菌群结构特征,初步探讨中药黄连解毒汤(HLJDD)干预作用影响。方法12周龄雄性SHR随机分模型组(M组)、HLJDD组(H组),同周龄雄性Wistar大鼠为正常对照组(W组),每组各4只。空白对照组及模型组常规饲养,HLJDD组按0.027 g/(kg·d)灌胃给药,每天给药1次,连续给药6周。干预结束后,取出盲肠内容物(新鲜粪便),采用16S rDNA高通量测序并进行生物信息学分析。结果3组大鼠肠道微生物测序覆盖度在0.997以上,从属水平分析3组大鼠物种丰度聚类分布于4个优势的细菌门:27个属于厚壁菌门类(Firmicutes)、4个属于拟杆菌门类(Bacteroidetes)、3个属于变形菌门类(Proteobacteria)和1个属于疣微菌门类(Verrucomicrobia)门类细菌;Simpson指数表明,模型组大鼠肠道菌群多样性降低,HLJDD组大鼠肠道菌群多样性高于模型组;3组大鼠肠道菌群微生物群落结构在门水平上主要由厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)和变形菌门(Proteobacteria)组成,其中模型组厚壁菌门(Firmicutes)相对丰度增加,HLJDD组大鼠肠道菌群厚壁菌门(Firmicutes)的相对丰度低于模型组;3组大鼠肠道菌群微生物群落结构在属水平上主要由乳酸杆菌属(Lactobacillus)、肠球菌属(Enterococcus)、大肠志贺氏杆菌属(Escherichia Shigella)组成,其中益生菌乳酸杆菌属(Lactobacillus)相对丰度在模型组明显降低,HLJDD组大鼠肠道菌群乳酸杆菌属相对丰度高于模型组。结论SHR肠道菌群结构失于平衡状态,菌群多样性有一定程度下降,HLJDD可能通过增加SHR菌群多样性、降低SHR致病菌厚壁菌门(Firmicutes)的相对丰度、升高SHR益生菌乳酸杆菌属相对丰度达到对抗高血压的作用,尚需扩大样本量进一步进行验证。Objective To analyze the structure of intestinal flora in spontaneous hypertension rats(SHR)anddiscuss the effect of Huanglian Jiedu Decoction(HLJDD).Methods Twelve weeks old male SHR were randomly divided into model group(M group)and HLJDD group(H group),and Wistar male rats of the same age were as the normal control group(W group),4 rats in each group.The normalcontrol group and model group were fed normally.HLJDD group was given 0.027 g/(kg·d)by gavage for 6 weeks.After the intervention,the cecal contents(fresh feces)were taken out,and 16 S rDNA high-throughput sequencing was used for bioinformatics analysis.Results The three groups’gut microbes sequencing coverage was above 0.997.The subordinate level analysis of three groups’species abundance distribution of clustering of rats in four advantages of bacteria gate:27 belonging to Firmicutes,4 belonging to Bacteroidetes,3 belonging toProteobacteria and 1 belonging to Verrucomicrobia.Simpson index showed that the diversity of intestinal flora in model group decreased,while that in HLJDD group was higher than that in model group.At the phylum level,the microbial community structure of the intestinal flora of the three groupswas mainly composed of Firmicutes,Bacteroidetes and Proteobacteria.The relative abundance of Firmicutes in model group was increased,while that in HLJDD group was lower than that in model group.The microflora structure of the intestinal flora of the three groups was mainly composed of Lactobacillus,Enterococcus and Escherichia Shigella at the genus level.The relative abundance of Lactobacillus was significantly reduced in the model group,and the relative abundance of Lactobacillus in the intestinal flora of rats in the HLJDD group was higher than that in the model group.Conclusion SHR intestinal flora structure has an equilibrium state and the flora diversity has a certain degree of decline.HLJDD may relive high blood pressure by increasing the intestinal flora of SHR,reducingthe relative abundance of Firmicutes,increasing the relative ab
