马钱子碱对人结肠癌细胞SW480增殖和周期的影响及机制研究

摘　　要：目的探讨马钱子碱对人结肠癌细胞株SW480的增殖和周期的影响及可能的分子机制。方法将试验分为空白对照组和马钱子碱高(8μmol/L)、中(4μmol/L)、低剂量组(2μmol/L),细胞生长曲线检测马钱子碱干预前后各组SW480细胞的增殖情况;流式细胞术检测马钱子碱对各组SW480细胞周期的影响;RT-PCR检测马钱子碱对SW480细胞中Wnt3a、β-catenin、C-myc、CyclinD1的mRNA表达的调节作用;Western Blot试验检测马钱子碱对SW480细胞中Wnt3a、β-catenin、C-myc、CyclinD1蛋白表达的调节作用。结果细胞生长曲线表明,在一定范围内,马钱子碱给药浓度越高,对SW480细胞增殖的抑制作用越显著,以高剂量组为最优。流式细胞术试验表明,马钱子碱可显著升高G2/M期的SW480细胞数量,从而将SW480周期阻滞在G2/M期。RT-PCR和Western Blot试验结果显示,马钱子碱干预后Wnt3a、β-catenin、C-myc、CyclinD1的mRNA和蛋白表达相较于正常对照组其表达明显降低。结论马钱子碱可通过调节Wnt/β-catenin信号通路抑制结肠癌细胞SW480增殖并将SW480细胞周期阻滞在G2/M期。Objective To investigate the effects and possible molecular mechanism of brucine on the proliferation and cycle of human colon cancer cell line SW480.Methods The experiment was divided into control group and brucine high(8μmol/L),medium(4μmol/L),low(2μmol/L)dose groups.Cell growth curve was used to detect the proliferation of SW480 cells in each group before and after intervention with strychnine brucine.Flow cytometry was used to detect the effect of brucine on the cell cycle of SW480 in each group.RT-PCR was used to detect the regulation of the expressions of Wnt3 a,β-catenin,C-myc and CyclinD1 in SW480 cells by brucine.Western Blot assay was used to detect the regulation of the expressions of Wnt3 a,β-catenin,C-myc and CyclinD1 in SW480 cells by brucine.Results The cell growth curve indicated that the higher the concentration of brucine administered,the more significant inhibition of SW480 cell proliferation was in a certain range,and the high dose group was the best.Flow cytometry experiments showed that brucine significantly increased the number of SW480 cells in the G2/M phase,and thus blocking the SW480 cycle in the G2/M phase.The results of RT-PCR and Western Blot showed that the mRNA and protein expressions of Wnt3 a,β-catenin,C-myc and CyclinD1 were significantly lower than those in the normal control group after intervention with brucine.Conclusion Brucine can inhibit the proliferation of colon cancer cell SW480 and arrest the SW480 cell cycle in G2/M phase by regulating Wnt/β-catenin signaling.
