摘 要:目的通过观察前列消汤对实验性自身免疫性前列腺炎(EAP)(湿热证)小鼠模型前列腺组织核转录因子-κB(NF-κB)P65和P50表达的影响,探讨前列消汤干预EAP模型前列腺内NF-κB的作用机制。方法建立EAP(湿热证)模型,用Western Blot和荧光定量PCR法、免疫组化法检测前列消汤对NF-κB P65和P50表达的影响。结果Western Blot结果显示,模型组与正常组比较,前列腺组织内NF-κB P65和P50的表达显著升高(P<0.05);与模型组比较,前列消高、中剂量组NF-κB P65的表达显著下降(P<0.05),前列消高、低剂量组NF-κB P50的表达显著下降(P<0.05)。荧光定量PCR结果显示,模型组与正常组比较,前列腺组织内NF-κB P65和P50的表达显著升高(P<0.05);与模型组比较,前列消高、中、低剂量组NF-κB P65的表达显著下降(P<0.05),前列消高剂量组NF-κB P50的表达显著下降(P<0.05)。免疫组化法显示,模型组与正常组比较,前列腺组织内NF-κB P65的表达显著升高(P<0.05);与模型组比较,前列消高、中剂量组NF-κB P65的表达显著下降(P<0.05)。结论前列消汤可通过抑制前列腺组织内NF-κB P65和P50的表达在CP的治疗中发挥作用。Objective To investigate the effect of Qianliexiao(QLX)Decoction on the expressions of prostate tissues NF-κB P65 and P50 in EAP(damp-heat syndrome)model mice and explore its mechanisms.Methods We built a EAP(damp-heat syndrome)model and used the expressionsof NF-κB P65 and P50 treated with QLX Decoction detected by Western Blot and fluorescence quantitative PCR,immunohi stochemistry.Results Western Blot showed that the expressions of NF-κB P65 and P50 in model group were higher than those of the normal group and the difference was statistically significant(P<0.05).Compared with the model group,the expressions of NF-κB P65 in high and middle dose groups were lower than those of the model group and the difference was statistically significant(P<0.05).The expressions of NF-κB P50 in highand low dose groups were lower than those of the model group and the difference was statistically significant(P<0.05).The fluorescence quantitative PCR showed that the expressions of NF-κB P65 and P50 in model group were higher than those of the normal group and the difference was statistically significant(P<0.05).Compared with the model group,the expressions of NF-κB P65 in high,middle and low dose groups were lower than those of the model group and the difference was statistically significant(P<0.05).The expressions of NF-κB P50 in high dose group were lower than those of the model group and the difference was statistically significant(P<0.05).The immunohistochemistry showed that the expressions of NF-κB P65 in model group were higher than those of the normal group and the difference was statistically significant(P<0.05).However,NF-κB P65 in high and middle dose groups were lower than those ofthemodel group and the difference was statistically significant(P<0.05).Conclusion QLX Decoction works through inhibiting the expressions of NF-κB P65 and P50 in prostate tissuetotreat chronic prostatitis.
