摘  要:目的比较脱皮马勃不同药用以及极性部位提取物对小鼠的止血作用、对亚甲基蓝吸附力的效果,为马勃的加工炮制方法提供实验依据。方法取脱皮马勃,剪成小块,分出孢体、包被,分别制成水煎液;对脱皮马勃45%乙醇提取液分别用石油醚、二氯甲烷、乙酸乙酯和正丁醇进行萃取,得到石油醚、乙酸乙酯、正丁醇和水层4个极性部位,将上述水煎液及极性部位作为不同组别的给药药物。采用断尾法、毛细管法测定出血时间(BT)、凝血时间(CT);血凝仪法测定血浆凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)和纤维蛋白原(FIB)含量;使用One-way ANOVA处理数据;利用分光光度法测定孢体、包被对亚甲基蓝的吸附力。结果与空白组比较,孢体组和包被组均可显著缩短小鼠的BT、CT、PT和APTT(PSymbol|@@0.05),孢体组可极显著提高FIB的含量(PSymbol|@@0.01),包被组显著缩短TT(PSymbol|@@0.05);脱皮马勃乙酸乙酯和正丁醇部位组均可显著缩短小鼠的BT、CT、PT、APTT、TT,提高其FIB的含量(PSymbol|@@0.05,PSymbol|@@0.01);对亚甲基蓝的吸附力(mg/g):包被5.744>孢体5.479。结论脱皮马勃的孢体、包被,其萃取物乙酸乙酯和正丁醇部位均有显著的止血作用,凝血机制可能与通过介导内源性、外源性凝血途径有关,且包被的吸附力大于孢体,建议统一将包被作为药用部位,规范马勃的加工炮制方法。Objective To compare the hemostatic effect of different medicinal and polar extracts of Lasiosphaera fenzlii Reich.on mice and the effect on the adsorption force of methylene blue,so as to provide experimental basis for the processing method of Lasiosphaera fenzlii Reich.Methods The Lasiosphaera fenzlii Reich.was cut into small pieces,and then the sporophyte and peridium were separated to make the decoction respectively.And then we extracted 45%ethanol extract from Lasiosphaera fenzlii Reich.with petroleum ether,dichloromethane,ethyl acetate and n-butanol respectively.The four polar parts of petroleum ether,ethyl acetate,n-butanol and water extraction sites were obtained,and the above-mentioned decoction and polar sites were used as different groups of drugs.Bleeding time(BT)and clotting time(CT)were determined by tail-breaking method and glass capillary tube method.Plasma prothrombin time(PT),activated partial thromboplastin time(APTT),thrombin time(TT),and fibrinogen(FIB)were measured by blood coagulation analyzer.The data was processed with One-way ANOVA.The adsorption force of sporophyte and peridium on methylene blue was determined by spectrophotometry.Results Compared with the blank group,the sporophyte group and the peridium group could significantly shorten the BT,CT,PT and APTT(P<0.05),and the sporophyte group could significantly increase the FIB content(P<0.01).The peridium group shortened TT(P<0.05)and the ethyl acetate group and the n-butanol group significantly shortened BT,CT,PT,APTT and TT of mice and increased the content of FIB(P<0.05,P<0.01).The adsorption force for methylene blue was peridium(5.744 mg/g)>sporophyte(5.479 mg/g).Conclusion The sporophyte and peridium of Lasiosphaera fenzlii Reich.have significant hemostatic effect and are the same to the ethyl acetate and n-butanol fractions.The coagulation mechanism may be related to the endogenous and exogenous coagulation pathways.And the adsorption force of peridium is greater than that of the sporophyte.It is recommended that the peridium sh
