摘 要:目的探讨雷公藤多苷片(TW)对糖尿病肾病(DN)大鼠肾组织RhoA/ROCK1表达的影响。方法SD雄性大鼠随机分为空白组、模型组、TW组。空白组和模型组灌服生理盐水,TW组给予TW片。8周后测体质量、肾质量、肥大指数及肾小球体积;生化检测24 h尿蛋白、血清白蛋白(ALB)、血脂(TC、TG)、空腹血糖(FBG)等;过碘酸-雪夫氏(PAS)染色观察肾脏病理;酶联免疫吸附测定(ELISA)法检测肾组织RhoA、ROCK1的浓度值。结果模型组ALB减少,体质量、肥大指数、TG、TC及FBG升高,肾小球体积增大,RhoA、ROCK1的表达增多,与空白组比较(P<0.05或P<0.01),具有统计学差异,说明造模成功。TW干预后,RhoA、ROCK1的表达下调,24 h尿蛋白减少,ALB升高,血脂、血糖及肥大指数降低,肾小球直径缩小,大鼠体质量及肾脏病理改善,足细胞损害减轻与模型组比较(P<0.05或P<0.01),具有统计学意义。结论TW可能通过影响DN大鼠肾组织RhoA、ROCK1的表达,而改善上述生化指标,减轻肾脏病理损害,且对肝、肾功能未见明显损害。Objective To investigate the effects of tripeygium wilfordi multiglucoside tablets on the expressions of RhoA and Rock1 in renal tissue of rats with diabetic nephropathy.Methods SD rats were randomly divided in to 3 groups:blank group,model group and TW group.The blank group and model group were given normal saline by gavage.TW group were given TW by gavage.The weight of rat,weight of kidney,hypertrophy index and the glomerular volume were measured eight weeks later.In addition,some indicators such as the quantity of 24 h uninary protein,serum albumin(ALB),serum total cholesterol(TC),triglyceride(TG)and fasting blood glucose(FBG)were determined by means of biochemical analysis eight weeks later.Renal pathology was observed by periodic acid-schiefs reaction(PAS).The enzyme linked immunosorbent assay(ELISA)was used to measure the levels of RhoA and ROCK1 in rat renal tissue.Results TW could down-regulate the expressions of RhoA and Rock1 and increase the 24 h urinary protein.TW could increase the level of albumin(ALB)and decrease the blood liquid,blood glucose and hypertrophy index.TW could also shorten the glomerular diameter and improve the weight of the rats and the renal pathology and could reduce the damage of podocytes.Conclusion TW could regulate the expressions of RhoA and Rock1 in renal tissue of rats with diabetic nephropathy,which is likely to improve some biochemical indicators related to renal pathological damage.In addition,there is no obvious damage to liver and kidney function.
