摘 要:目的研究补气活血药物黄芪、莪术配伍对斑马鱼血管生成数量和结构的影响。方法将不同浓度的黄芪、莪术1∶1配伍水煎剂与斑马鱼胚胎共同孵育24 h,采用立体显微镜成像统计体节间血管根数和长度,采用激光共聚焦成像统计体节间血管面积,并观察体节间血管的结构和顶端细胞丝状伪足的伸展。结果与空白对照DMSO组相比,黄芪、莪术配伍组体节间血管根数、长度、面积均减少(P<0.05或P<0.01),且黄芪、莪术配伍高剂量组抑制作用最强。黄芪、莪术配伍组斑马鱼体节间血管变短,甚至缺失,同时管壁完整,形态规则,走行自然,均匀等距。结论补气活血药物黄芪、莪术配伍不仅通过抑制顶端细胞丝状伪足的伸展抑制斑马鱼血管生成,而且保持血管形态均匀规则。斑马鱼可以作为评价中药抗血管生成活性的整体实验模型,为深入研究补气活血药物改善肿瘤血管的形态结构提供了方向。Objective to observe the effect of the compatibility of Huangqi(radix astragali)and Ezhu(curcuma)on the number and structure of angiogenesis in zebrafish.Methods The equal proportion of Huangqi(radix astragali)and Ezhu(curcuma)was incubated with zebrafish embryos for 24 h.Stereotactic microscopy was used to measure the numbers and length of intersegmental blood vessels.Laser confocal imaging was used to count the areas of intersegmental blood vessels.Besides,the structure of intersegmental blood vessels and the filopodia extension of top cells were observed.Results Compared with the DMSO group,the numbers,length and areas of intersegmental blood vessels were inhibited in compatibility group(P<0.05 or P<0.01).High dose group had the strongest inhibitory effect.The length of intersegmental blood vessels was shortened or even missing by the compatibility of Huangqi(radix astragali)and Ezhu(curcuma).The vessel wall was intact and the morphology was regular and equidistant.Conclusion The compatibility of Huangqi(radix astragali)and Ezhu(curcuma)not only inhibited the angiogenesis of zebrafish by inhibiting the filopodia extension of top cells,but also maintained the regularity of the vascular morphology.Zebrafish could be used as an overall experimental model to evaluate the anti-angiogenic activity of traditional Chinese medicine,which provided a direction for further research on the morphological structure of tumor blood vessels by reinforcing Qi and activating blood drugs.
