摘 要:旨在研究小麦TaGRF4基因的生物学功能,利用生物信息学技术分析小麦TaGRF4基因结构及其与其他作物的进化关系,利用实时定量检测TaGRF4在不同发育时期叶片和不同区段叶片中的表达丰度,同时检测外源GA处理条件下TaGRF4的表达情况,最后利用BSMV-VIGS体系使TaGRF4的3个同源基因同时沉默,研究TaGRF4沉默对叶片生长发育的影响。结果表明,TaGRF4是玉米ZmGRF10的同源基因,在幼嫩叶片和叶尖部位的表达量最高,而且对外源GA处理响应迅速;TaGRF4沉默后新生叶片明显伸长,说明TaGRF4与ZmGRF10有类似的生物功能,能够抑制叶片的伸长生长。In order to investigate the biological function of wheat TaGRF4 gene,we first analyzed the gene structure of TaGRF4and its evolutionary relationship with other major crops by using bioinformatics methods,and then,the expression of TaGRF4 in leaves of different sections and different developmental stages were detected by real-time qPCR.The expression of TaGRF4 in leaves with GA treatment was also examined using real-time qPCR.after that,the BSMV-VIGS system was used to silence the expression of three TaGRF4 homoeologous genes simultaneously for studying the effect of TaGRF4 silencing on growth and development of leaves.The results indicated that TaGRF4 was a homologous gene of maize ZmGRF10,and mainly expressed in young leaves and leaf tips,moreover,the expression of TaGRF4 was induced by GA.The silencing TaGRF4 genes resulted in the elongation of new leaves,this indicated TaGRF4and ZmGRF10 had similar biological functionin the inhibition of leaf elongation.
