摘 要:目的探究蹄叶橐吾提取物(ethanolic extract of ligularia fischeri,EEOLF)的抗炎作用机制,为蹄叶橐吾(Ligularia fischeri,LF)的临床应用提供理论依据。方法采用脂多糖(lipopolysaccharide,LPS)作用于RAW264.7细胞建立体外细胞炎症模型。实验分6组,即空白对照组、EEOLF组、LPS组、LPS+吡咯烷二硫代氨基甲酸盐(Ammonium pyrrolidinedithiocarbamate,PDTC)组、LPS+EEOLF组、LPS+EEOLF+PDTC组。Western blot法检测药物作用于RAW264.7细胞后,细胞核内因子κB p65(nuclear factor-κB,NF-κB)的表达水平和磷酸化水平。ELISA法测定药物作用于RAW264.7细胞后IL-1β及TNF-α的释放量。结果与空白组对照比较,LPS能显著增加细胞核内NF-κB p65蛋白表达及其磷酸化水平(P<0.001);与LPS组比较,EEOLF能显著下调LPS诱导增加的NF-κB p65蛋白表达及其磷酸化水平(P<0.001)。与空白组比较,LPS能显著增加炎症因子L-1β及TNF-α的分泌(P<0.001);与LPS组比较,蹄叶橐吾提取物(ethanolic extract of ligularia fischeri,EEOLF)能显著降低LPS诱导增加的L-1β及TNF-α的分泌(P<0.001)。在NF-κB抑制剂PDTC作用下,LPS诱导的细胞炎症反应减弱,并且在PDTC存在下,EEOLF能更加明显降低LPS所诱导的细胞炎症反应。结论EEOLF通过作用于NF-κB通路抑制炎性因子的分泌,从而达到抗炎作用。To study the anti-inflammatory activity and molecular mechanism of ethanolic extract of ligularia fischeri(EEOLF),as to provide theoretical basis for the clinical application of Ligularia fischeri(LF).We used macrophage RAW264.7 cells induced by lipopolysaccharide(LPS)as an in vitro inflammatory model.PDTC was an NF-κB inhibitor.The experiment was divided into 6 groups,namely blank group,EEOLF group,LPS group,LPS+Ammonium pyrrolidinedithiocarbamate(PDTC)group,LPS+EEOLF group,LPS+EEOLF+PDTC group.Then western blot was used to explore the expression of NF-κB p65 and p-p65 in nucleus.ELISA was used to observe the relaese of IL-1βand TNF-α.Compared with the blank group,the expression of nuclear factor-κB p65(NF-κB p65)and its phosphorylation were significantly increased in the LPS group(P<0.001);compared with the LPS group,pretreatment with EEOLF significantly reduced the expression of NF-κB p65 and p-p65(P<0.001).Compared with the blank group,the secretion level of IL-1βand TNF-αwere significantly increased in the LPS group(P<0.001);compared with the LPS group,pretreatment with EEOLF significantly decreased the release of IL-1βand TNF-α(P<0.001).Under the effect of NF-κB inhibitor PDTC,the inflammatory response induced by LPS was weakened.And in the presence of PDTC,EEOLF could significantly reduce the inflammatory response induced by LPS.EEOLF can inhibit the secretion of inflammatory factors through the NF-κB pathway,thereby achieving anti-inflammatory effects.
