摘  要:目的通过观察填精通络方治疗糖尿病性勃起功能障碍大鼠勃起功能的疗效及其对RhoA/Rho信号通路的影响,探讨填精通络方在治疗糖尿病性勃起功能障碍的作用机制。方法将40只SD大鼠随机分为正常组、模型组、对照组和填精通络组,每组10只。除正常组外,其余各组以腹腔注射链脲佐菌素制备糖尿病大鼠模型,任意时间血糖>16.7 mmol/L为糖尿病成模标准,然后用阿朴吗啡阴茎勃起实验筛选糖尿病性勃起功能障碍大鼠模型。成模后,对照组予以他达拉非灌胃,填精通络组予以填精通络方灌胃,正常组与模型组分别灌服等体积蒸馏水,干预4周后评估各组大鼠的勃起功能,HE染色法观察阴茎海绵体组织的病理变化,并采用Real-time PCR、WB的方法分别检测阴茎海绵体组织中RhoA、ROCK1、ROCK2的mRNA及蛋白的表达。结果对照组和填精通络组阴茎勃起次数显着增加,潜伏期缩短(P<0.01和P<0.05)。与正常组相比,模型组、对照组和填精通络组RhoA,ROCK1和ROCK2的mRNA水平显着升高(P<0.01和P<0.05)。与模型组相比,对照组和填精通络组RhoA,ROCK1和ROCK2 mRNA的表达显着降低(P<0.01和P<0.05)。结论填精通络方可以改善糖尿病性勃起功能障碍大鼠的勃起功能,其具体机制可能是通过下调RhoA/Rho通路中的RhoA、ROCK1、ROCK2的mRNA及蛋白表达来实现的。Objective To investigate the treatment mechanism of Replenishing Essence and Dredging Collaterals Formula(REDCF)in treating erectile dysfunction by observation of REDCF’s efficacy in treating erectile function in diabetes-induced erectile dysfunction rats and its impact on the RhoA/Rho signaling pathway.Method Forty SD rats were randomly divided into four groups:normal group,model group,control group,and REDCF group.There were 10 animals in each group.Other than the normal group,the other groups were injected with streptozotocin intraperitoneally to create a diabetic rat model.Blood glucose>16.7 mmol/L at random was the standard for successful diabetic rat model.Apomorphine penile erection experiment was then used to screen rat models with diabetic erectile dysfunction.After the model was successfully made,the control group was given tadalafil for gavage.The TCM group was given gavage with REDCF.The normal group and the model group were orally administered with an equal volume of distilled water.After 4 weeks of drug intervention,the erectile function of each group of rats was evaluated.HE staining was used to observe the pathological changes of the corpus cavernosum tissue of rats in each group.Real-time PCR and WB methods were used to detect the expression of RhoA,ROCK1,ROCK2 mRNA and protein in penile cavernous tissue,respectively.Results The number of penile erections in the control group and the REDCF group increased significantly,and the latency period was shortened(P<0.01 and P<0.05).Compared with the normal group,the mRNA levels of RhoA,ROCK1,and ROCK2 in the model group,control group,and the REDCF group were significantly increased(P<0.01 and P<0.05).Compared with the model group,the levels of RhoA,ROCK1 and ROCK2 mRNA in the control group and the REDCF group were significantly reduced(P<0.01 and P<0.05).Conclusion Replenishing Essence and Dredging Collaterals Formula can improve erectile function in diabetes-induced erectile dysfunction rats.Its specific mechanism may be achieved by down-regulating th
