摘 要:目的基于磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)信号通路探究黄芪葛根配伍降低糖尿病大鼠血糖、血脂的作用机制。方法SD大鼠一次性腹腔注射链脲佐菌素(STZ)诱导糖尿病模型,随机分为模型组、阳性对照(金芪降糖片)组(1.47 g/kg)、黄芪组(2.7 g/kg)、葛根组(1.35 g/kg)、黄芪葛根汤组(4.05 g/kg),各15只,另设正常组11只,造模同日灌服相应药物或蒸馏水10 mL/kg,连续30 d。检测血糖、甘油三酯(TG)、胆固醇(CHO),实时荧光定量PCR法检测肝组织胰岛素受体底物-2(IRS-2)、磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT2)、糖原合成酶激酶3β(GSK3β)、肝X受体(LXRα)、固醇调节元件结合蛋白-1(SREBP-1)mRNA相对表达量。结果与正常组比较,模型组大鼠治疗30 d后血糖、CHO、TG水平明显升高,IRS-2、PI3K、AKT2、LXRαmRNA表达降低,GSK3β、SREBP-1 mRNA表达增加(P<0.05)。黄芪组、葛根组及其配伍组能降低模型组大鼠血糖及CHO、TG水平;提升IRS-2、PI3K(葛根组除外)、AKT2(葛根组除外)、LXRαmRNA表达,但两药间无交互作用;同时能降低GSK3β、SREBP-1 mRNA表达,黄芪、葛根二者配伍降低GSK3βmRNA表达的效果优于单用,而降低SREBP-1 mRNA表达的效果不如单用。结论黄芪降糖、降脂作用可能与提升IRS-2、PI3K/AKT、LXRαmRNA表达,降低GSK3β、SREBP-1 mRNA表达有关。葛根降糖、降脂作用可能与提升IRS-2、LXRαmRNA表达,降低GSK3β、SREBP-1 mRNA表达有关。黄芪葛根配伍降低糖尿病大鼠的血糖血脂可能与刺激肝组织IRS-2,激活PI3K/AKT信号通路,调节下游信号分子GSK3β、LXRα、SREBP-1的活性有关。Objective To explore the effects and mechanism of Radix Astragali-Pueraria Lobata compatibility on reducing blood glucose and lipids of diabetic rats based on PI3K/AKT signaling pathway.Methods SD rats were induced to diabetes model by intraperitoneal injection of streptozotocin(STZ)one time and randomly divided into model group,positive control(Jinqi Jiangtang)group(1.47 g/kg),Radix Astragali group(2.7 g/kg),Pueraria Lobata group(1.35 g/kg)and Radix Astragal-Pueraria Lobata compatibility group(4.05 g/kg),15 rats in each group,additional 11 rats of normal group.On the same day,rats were given corresponding drugs or distilled water(10 mL/kg)for 30 days.Glucose,triglyceride(TG)and cholesterol(CHO)in serum were detected by biochemical method.Real-time PCR was used to detect the relative expression of insulin receptor substrates-2(IRS-2),phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT2),glycogen synthase kinase 3β(GSK3β),liver X receptorα(LXRα)and sterol-regulatory element binding protein 1(SREBP-1)mRNA in rat liver tissues.Results Compared with the normal group,the level of blood glucose,TG and CHO were increased in model group,the expressions of IRS-2,PI3K,AKT2 and LXRαmRNA were decreased,the expressions of GSK3βand SREBP-1 mRNA were increased(P<0.05).Astragali Radix,Puerariae Lobata group and their compatibility group reduced the level of blood glucose,TG,CHO and improved the expression of IRS-2,PI3K(except Puerariae Lobata group),AKT2(except Puerariae Lobata group),LXRαmRNA,but there was no interaction between the two drugs,meanwhile,reduced the expression of GSK3β,SREBP-1 mRNA.The compatibility group showed better effect than single drug in reducing GSK3βmRNA,the inhibitory effect of each single drug on the expression of SREBP-1 mRNA was better than the compatibility group.Conclusion Radix Astragali can reduce blood glucose and lipids by improving the expression of IRS-2,PI3K/AKT,LXRαmRNA and reducing the expression of GSK3β,SREBP-1 mRNA.While Pueraria Lobata can reduce blood glucose and lipi
