摘  要:目的:探究β_2-肾上腺素能受体(ADRB2)基因多态性/单倍型对湖州地区支气管哮喘患者易感性及吸入沙美特罗替卡松的治疗效果的影响。方法:选取2015年12月-2018年3月在浙江省湖州市第一人民医院呼吸内科治疗的102例支气管哮喘急性发作期患者为哮喘组,另选取105例健康体检者为对照组,采用TaqMan探针测定两组ADRB2基因编码区的16、27位点基因多态性,分析单倍型。哮喘组予沙美特罗替卡松粉吸入剂(50 μg/250 μg)吸入治疗,2次/d,共治疗16周,比较治疗前后肺功能的改善情况。结果:两组16、27位点各基因及单倍型频率分布比较,差异均无统计学意义(P>0.05)。哮喘组ADRB2各基因型ΔFEV_1%、ΔFVC%及ΔPEF%比较,差异均无统计学意义(P>0.05)。结论:ADRB2基因16、27位点多态性/单倍型频率分布在哮喘患者与健康者中无区别,ADRB2基因多态性/单倍型不影响支气管哮喘患者应用沙美特罗替卡松治疗的反应性。Objective:To explore the susceptibility of the beta-2 adrenergic receptor(ADRB2)gene polymorphism/haplotype to bronchial asthma in Huzhou,and its influence on therapeutic effect of Salmeterol Xinafoate and Fluticasone Propionate.Method:A total of 102 patients at the acute stage of bronchial asthma treated in the department of respiratory medicine of The First People’s Hospital of Huzhou,Zhejiang Province from December 2015 to March 2018 were selected as the asthma group,and another 105 healthy subjects were selected as the control group.The polymorphisms of 16 and 27 locis in the coding region ADRB2 gene in two groups were determined by the TaqMan probe and the haplotypes were also analyzed.The asthma group was treated with Salmeterol Xinafoate and Fluticasone Propionate Powder for Inhalation(50μg/250μg),twice a day,and treatment lasted 16 weeks.The improvement of pulmonary function before and after treatment were compared.Result:The frequency distribution of each gene and haplotype at 16 and 27 locis were compared,and the differences were not statistically significant(P>0.05).In the asthma group,ΔFEV1%,ΔFVC%andΔPEF%of each gene of ADRB2 were compared,and the differences were not statistically significant(P>0.05).Conclusion:There are no significant differences in the frequency distribution of ADRB2 gene polymorphisms/haplotypes at 16 and 27 locis between asthmatic patients and healthy subjects.ADRB2 gene polymorphisms/haplotypes don’t affect the response of patients with bronchial asthma to treatment with Salmeterol Xinafoate and Fluticasone Propionate.
