摘 要:目的观察鸡豆黄素A(Bioch A)对1-甲基-4-苯基吡啶离子(MPP^+)诱导的PC12细胞的凋亡是否有保护作用。方法用MTT法检测MPP^+对PC12细胞的毒性作用和Bioch A的最适药物浓度,为后续实验筛出诱导细胞凋亡的MPP^+剂量和恰当的Bioch A剂量。常规培养的PC12细胞被分为:Control组、MPP^+(1μmol/L)组、Bioch A(1.25μmol/L)+MPP^+组、Bioch A(2.5μmol/L)+MPP^+组、Bioch A(5μmol/L)+MPP^+组、雌二醇(E2)(0.1μmol/L)+MPP^+组。DCFH-DA探针被用于活性氧(ROS)的检测;Annexin V-FITC/PI双染法被用作细胞凋亡百分比的测定;cleaved Caspase-3和cleaved Caspase-9的表达水平采用Western blot法检测。结果MPP^+模型组较Control组其ROS含量有所升高,Bioch A(2.5、5μmol/L)组的ROS的含量有所下降。MPP^+模型组与Control组比较,其细胞凋亡百分率和活化的cleaved Caspase-3以及cleaved Caspase-9蛋白表达量增多,Bioch A(2.5、5μmol/L)组与MPP^+模型组比较,细胞凋亡百分率下降,cleaved Caspase-3和cleaved Caspase 9蛋白的表达量减少。结论Bioch A对MPP^+诱导的PC12细胞凋亡具有一定的保护作用。Objective To observe whether biochanin A(Bioch A)exerts protective effects on MPP^+-induced PC12 cells apoptosis.Methods Effects of MPP^+and Bioch A on PC12 cells viability were detected by MTT assay to screen out the MPP^+-induced apoptosis dose and appropriate Bioch A dose.Conventional cultured PC12 cells were divided into:Control group,MPP^+(1μmol/L)group,Bioch A(1.25μmol/L)group,Bioch A(2.5μmol/L)group,Bioch A(5μmol/L)group and Estradiol(E2)(0.1μmol/L)group.Reactive oxygen species(ROS)was detected by DCFH-DA probe method.Apoptosis rate was detected by Annexin V-FITC/PI double staining.The expression levels of cleaved Caspase-3 and cleaved Caspase-9 were detected by Western blot method.Results Compared with the control group,ROS content increased in MPP^+group.Compared with the MPP^+group,ROS content decreased in Bioch A(2.5,5μmol/L)groups.Compared with the control group,the apoptosis rate and the levels of cleaved Caspase-3 and cleaved Caspase-9 increased in MPP^+group.Compared with the MPP^+group,the rate of apoptosis and the expression levels of cleaved Caspase-3 and cleaved Caspase-9 decreased in Bioch A(2.5,5μmol/L)groups.Conclusion Bioch A exerts protective effects on MPP^+-induced PC12 cells apoptosis.
