基于亲权鉴定技术探究种鸡纯繁的合适停输间隔

摘　　要：试验旨在探究新输入精液能否完全取代母鸡储精腺残留精液,为种鸡纯系继代繁育时合适的停输间隔提供依据。选择白来航母鸡40只分为A、B两组,公鸡8只分为G1、G2两组。第一次对A、B两组母鸡分别输G1、G2组公鸡精液;10 d后进行第二次输精,两组公鸡交换。首次输精后两天开始收集种蛋,共30 d。提取孵化4 d的鸡胚组织和亲本血液DNA。选用13对微卫星标记对后代和亲代的DNA样本进行PCR扩增和毛细血管电泳,再通过GeneMarker识别样本的基因型,利用Cervus对鸡胚进行父本鉴定。结果显示:10 d输精间隔条件下,第二次输精的公鸡后代占据主导地位,但第二次输精并不能完全取代第一次输精,第一次输入的精液可在母鸡输卵管内储存长达25 d,且维持受精潜能。研究认为种鸡纯繁时,为保证系谱的准确性,停输时长在25 d以上较为合理,且纯繁过程中尽量避免更换公鸡。The objective of the experiment was to investigate whether the newly semen could completely replace the residual spermatozoa which stored in sperm storage tubes of hens.Our findings will provide foundation to determine the appropriate insemination interval in chicken purebred reproduction.Forty White Leghorns hens were equally divided into two groups(A and B).Eight roosters were divided into two groups(G1 and G2).Hens in groups A and B were inseminated with G1 and G2,respectively.The second insemination were performed in 10 days later,and hens in groups A and B were inseminated with G2 and G1,respectively.Eggs were collected daily after the first insemination for30 days.DNA were extracted from parental blood and embryos that had incubated to day 4.A total of 13 microsatellite primer pairs were employed for PCR amplification.The amplification products were separated by capillary electrophoresis.Genotype of each loci were identified using GeneMarker software,and the parentage analysis were conducted with Cervus software.The results showed that the sperm from the latest insemination could not completely replace the sperm that stored in female reproductive tract,although the paternity of offspring were dominated by the most recent inseminating sperm.The sperm could store in hen′s reproductive tract for up to 25 days and maintained potential fertilizing ability.In order to ensure the accuracy of the pedigree in chicken purebred reproduction,it is necessary to keep the insemination interval for more than 25 days and avoid replacing the rooster during breeding.
