摘 要:目的探讨蛹虫草多糖灌胃对氟尿嘧啶(5-FU)处理大鼠骨髓功能的影响。方法水溶醇沉法分离蛹虫草多糖。将40只雄性Wistar大鼠随机分为4组,分别为对照组、模型组、蛹虫草多糖低剂量组、蛹虫草多糖高剂量组,每组10只。除对照组外,其余各组腹腔注射15 mg/(kg·d)5-FU,连续6 d。蛹虫草多糖低、高剂量组分别给予20、60 mg/kg的蛹虫草多糖粗品灌胃,3次/d,连续给药3周,其余组给予同等体积的生理盐水。观察大鼠体质量变化,采用全自动生化仪测定外周血细胞[红细胞(RBC)、白细胞(WBC)、血小板(PLT)、血红蛋白(Hb)]含量,采用ELISA法测定血清IFN-γ和IL-4,采用酶标仪测定骨髓DNA含量。结果与对照组相比,模型组体质量、RBC、WBC、PLT、Hb、IFN-γ/IL-4(Th1/Th2)、骨髓DNA含量均降低(P均<0.01)。与模型组相比,蛹虫草多糖低、高剂量组大鼠体质量差异无统计学意义,蛹虫草多糖高剂量组RBC、WBC、PLT、Hb、IFN-γ/IL-4(Th1/Th2)、骨髓DNA含量升高(P均<0.05)。结论蛹虫草多糖灌胃能有效减少5-FU所致的大鼠骨髓功能损伤,降低其毒副作用。Objective To investigate the effect of intragastric administration of Cordyceps militaris polysaccharide on bone marrow function of rats treated with 5-fluorouracil(5-FU).Methods We separated the Cordyceps militaris polysaccharide by water dissolution and alcohol precipitation.Forty Wistar male rats were randomly divided into 4 groups:the control group,model group,low-dose Cordyceps militaris polysaccharide group,and high-dose Cordyceps militaris polysaccharide group,with 10 rats in each group.In addition to the control group,rats in the other groups were intraperitoneally injected with 15 mg/(kg·d)5-FU for 6 consecutive days.The rats in the low-dose and high-dose Cordyceps militaris polysaccharide groups were given 20 and 60 mg/kg Cordyceps militaris polysaccharide crude by gavage,3 times a day for 3 consecutive weeks,and the remaining groups were given the same volume of normal saline.We observed the changes in weight of rats,and measured the content of peripheral blood cells[red blood cells(RBC),white blood cells(WBC),platelets(PLT),and hemoglobin(Hb)]by automatic biochemical analyzer,and detected the levels of serum IFN-γand IL-4 by ELISA,and measured the DNA content of bone marrow by Microplate reade.Results Compared with the control group,the body weight,RBC,WBC,PLT,Hb,IFN-γ/IL-4(Th1/Th2)and bone marrow DNA content all decreased in the model group(all P<0.01).There was no statistically significant difference in the body weight of rats between the model group and low-dose and high-dose Cordyceps militaris polysaccharide groups.In the high-dose Cordyceps militaris polysaccharide group,RBC,WBC,PLT,Hb,IFN-γ/IL-4(Th1/Th2)and DNA content of bone marrow increased(all P<0.05).Conclusion Cordyceps militaris polysaccharide can effectively alleviate the 5-FU-induced bone marrow function damage and reduce its toxic and side effects.
