摘　　要：【目的】探究不同时间点短链脂肪酸(SCFAs)对奶牛瘤胃上皮细胞促炎因子、趋化因子和紧密连接蛋白表达量的影响.【方法】试验分4组,每组设3个重复,分别在各组瘤胃上皮细胞中添加20 mmol/L的短链脂肪酸,在添加短链脂肪酸0、2、5、8 h后收集细胞提取细胞总RNA.【结果】在添加SCFAs 8 h时,G-蛋白偶联受体41(GPR41)的表达量显著高于其它各组(P<0.05).添加SCFAs 2 h后IL-1β的表达量显著高于0 h组(P<0.05),8 h时IL-1β的表达量最高.添加SCFAs 5、8 h时,CXCL2的表达量显著高于0 h组(P<0.05);在培养2 h后CXCL8的表达量显著高于0 h组(P<0.05).添加SCFAs 2 h后,Claudin-1和Occludin的表达量显著高于0 h组,并且在2 h时表达量最高(P<0.05).【结论】短链脂肪酸能够提高瘤胃上皮细胞促炎因子、免疫趋化因子、紧密连接蛋白以及GPR41的表达量,从而证明短链脂肪酸可以增强瘤胃上皮的免疫机能.【Objective】To evaluate the effects of short chain fatty acids(SCFA)on the expression of proinflammatory cytokine,chemokine and tight junction protein in bovine rumen epithelial cells at different time points.【Method】The experiment was divided into four groups,every group contained three replicates.The bovine rumen epithelial cells were incubated with 20 mmol/L SCFAs.After adding the short chain fatty acids 0,2,5,8 h,cells were collected to extract the total RNA.【Result】Compared with other groups,the GPR41 expression was significantly higher after adding SCFAs for 8 h(P<0.05).The expression of IL-1βwas significantly higher than that of 0 h group after adding SCFAs for 2 h(P<0.05),and reached the highest level at 8 h.The expression of CXCL2 was significantly higher than that of 0 h group at 5 and 8 h(P<0.05).The expression of CXCL8 was significantly higher than that of the 0 h group after 2 h of culture(P<0.05).The expression of Claudin-1 and Occludin was significantly higher than that of the 0 h group after adding SCFAs for 2 h,and reached the highest level at 2 h(P<0.05).【Conclusion】SCFAs could significantly enhance the expressions of proinflammatory cytokine,immunochemokine,tight junction protein and GPR41 in bovine rumen epithelial cells,which proves that an appropriate amount of short chain fatty acids can indeed enhance the immune function of rumen epithelium.
